Lane 1 : Wild type HAP1 whole cell lysate (20 μg)
Lane 2 : P Glycoprotein knockout HAP1 whole cell lysate (20 μg)
Lane 3 : HepG2 whole cell lysate (20 μg)
Lanes 1 - 3 : Merged signal (red and green). Green - ab170904 observed at 160 kDa. Red - loading control, ab8245, observed at 37 kDa.
ab170904 was shown to specifically react with P Glycoprotein when P Glycoprotein knockout samples were used. Wild-type and P Glycoprotein knockout samples were subjected to SDS-PAGE. ab170904 and ab8245 (Mouse anti GAPDH loading control) were incubated overnight at 4°C at 1 ug/ml and 1/10000 dilution respectively.
Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preabsorbed ab216773 and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed ab216776 secondary antibodies at 1/10000 dilution for 1 hour at room temperature before imaging.

Immunohistochemical analysis of formalin fixed paraffin embedded human kidney labelling P Glycoprotein with ab170904 at a concentration of 1µg/ml. The immunostaining was performed on a Ventana DISCOVERY ULTRA (Roche Tissue Diagnostics) instrument with a OptiView DAB IHC Detection Kit. Heat mediated antigen retrieval was performed with DISCOVERY cell conditioning solution (CC1) 100°C, pH8.5 for 32mins. ab170904 Anti-P Glycoprotein antibody [EPR10364-57] was incubated for 16mins at 37°C. Sections were counterstained with Hematoxylin II. Image inset shows absence of staining in secondary antibody only control. Customers are encouraged to optimise antigen retrieval conditions, antibody concentration, incubation times and temperature for best results in their own IHC assay workflow (automated and manual).

Biological function summary
P-glycoprotein acts as a protective mechanism against toxic substances and drugs within the body. It is a member of the ATP-binding cassette (ABC) transporter family which is critical for their ability to transport molecules across membranes. P-glycoprotein is not typically part of a larger protein complex but can interact with other membrane proteins to facilitate its function. Pathways
P-glycoprotein plays a significant role in the pharmacokinetics of drugs within the body. This transporter is involved in the multi-drug resistance (MDR) pathway which impacts the efficacy of chemotherapeutic agents.
