Anti-Ovalbumin抗体[6C8] (ab17293)

The two articles at the PubMed addresses below discuss the rationale for the preparation of the immunogen that generated the monoclonal antibody ab17293. The epitope that it detects (LEPINF, aa130-135) is listed in Table 1 of the 2014 paper, which refers to the clone as HYB 94-06.

Notes from the 2014 paper about the immunogen prep:

Formaldehyde-lysine (FK)-treated OVA was prepared by dissolving OVA in phosphate buffer followed by addition of 35% formaldehyde to a concentration of 0.035 M and lysine to a concentration of 0.025 M. The solution incubated at 35C with end-over-end agitation for 2 weeks. Following incubation, the solution was centrifuged (10 000 g for 30 min) and dialyzed against PBS. FK treatment is a well-known method for inactivation of toxins in vaccine development, where the proteins gain an acidic net charge, but retain their immunogenic and antigenic structure, which upon vaccination elicits protective Abs. The overall negative charge of FK-reacted proteins prevents aggregation and precipitation of the proteins as a result of heat treatment.

From the abstract of the 1996 paper:

Intraperitoneal immunization of rabbits and mice showed that both native and denatured forms of ovalbumin induced an immune response, but denatured forms of ovalbumin were found to be less immunogenic and to have a lower epitope density than native ovalbumin.

We have not had any complaint about this antibody since 2008. According to our record, we have sold several vials of this batch without any problem. This implies that it is very unlikely that the problem is batch-related.

Currently, we are not having a new batch from the Lab until this current batch is sold out completely.

We do recommend that you store ab17293 at +4C in the dark. Generally, antibodies stored at +4C are viable for up to a year. I wouldn't recommend freezing it. Since we haven't tested the antibody after it's been frozen, we do not know whether or not this would affect the antibody stability plus the antibody lacks any cryoprotectant.

The lab confirmed that the epitope that the antibody recognizes was not mapped. therefore I am unable to answer your question, if the epitope for this antibody is repeated in the protein and if it possible that this antibody binds the protein in more than one place.
