Immunocytochemistry/ Immunofluorescence - Anti-OTUD3 Antibody (AB316951)
Immunofluorescence staining of U251 (Human brain glioma cell line) cells with ab316951 at 1/50, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488® conjugated AffiniPure Goat Anti-Rabbit IgG(H+L).

Biological function summary
OTUD3 is essential in regulating protein homeostasis and cellular signaling. It is part of protein complexes that modulate ubiquitination pathways maintaining proper protein turnover and function. By deubiquitinating target proteins OTUD3 can influence processes like cell cycle progression apoptosis and DNA repair. The enzyme's regulation of these pathways highlights its role in maintaining cellular health.
Pathways
OTUD3 influences key signaling cascades such as the NF-kB and p53 pathways. In the NF-kB pathway OTUD3 interacts with proteins like TRAF3 modulating inflammation and immune responses. Similarly in the p53 pathway it can impact the stability and activity of p53 protein which is critical for cell cycle regulation and apoptosis. The relationship of OTUD3 with these pathways indicates its importance in cell signaling and stress responses.
