补充信息

Orexin Receptor 1 and 2 also known as hypocretin receptor 1 and 2 are G-protein-coupled receptors that bind to neuropeptides orexin-A and orexin-B. Orexin Receptor 1 has a molecular mass of approximately 50-60 kDa while Orexin Receptor 2 is similar in size. These receptors are mainly expressed in the brain particularly in the hypothalamus and other regions regulating wakefulness and sleep. The binding of orexins to these receptors triggers intracellular signaling cascades important for various physiological functions.
Biological function summary

These receptors play a significant role in modulating sleep-wake cycles and energy homeostasis. Orexin Receptors do not form part of a larger complex but interact with other proteins within the cell signaling networks. They are involved in the regulation of feeding behavior arousal and reward systems. Activation of these receptors promotes wakefulness and increases energy expenditure affecting the central nervous system's balance.

Pathways

Orexin Receptors participate in the neuropeptide signaling pathway and the neuronal system pathway. They interact with G-proteins to mediate their signals linking them to secondary messenger systems like cyclic AMP. These receptors also engage with proteins such as GABA receptors and adenosine receptors modulating inhibitory and excitatory neurotransmitter release to maintain alertness and metabolic processes.

Orexin Receptors are closely associated with sleep disorders such as narcolepsy and insomnia. Narcolepsy often involves the loss of orexin-producing neurons leading to disruptions in normal sleep patterns. These receptors also have connections to metabolic disorders like obesity where dysregulation of orexin signaling can affect appetite and energy expenditure. The alteration of these signals further involves interactions with leptin and ghrelin highlighting the complex nature of energy balance and appetite regulation.
