Biological function summary
Protein Nogo-A restricts neuronal regeneration and plasticity which can be observed during development and in adult nervous system tissues. It is part of the Nogo-66 receptor complex which includes receptors like NgR1. The interaction with its receptor complex triggers RhoA signaling pathways leading to the inhibition of cytoskeletal dynamics that are necessary for axon extension. This inhibition tends to stabilize synaptic connections and helps in maintaining the structural integrity of the nervous system.
Pathways
Nogo-A impacts the signaling pathways that are involved in axonal guidance and neuronal regeneration. The protein participates prominently in the RhoA/ROCK pathway which is responsible for regulating cytoskeletal formation and dynamics. Nogo-A is also related to the p75 neurotrophin receptor pathway which influences cell survival and apoptosis. These pathways collectively contribute to the regulation of neural growth and plasticity by modulating actin cytoskeleton structure in response to extrinsic inhibitory signals.
