Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-NEAS antibody [D8B7] (AB11755)
IHC staining of purified ab11755 on formalin-fixed paraffin-embedded mouse brain tissue. Following antigen retrieval using Sodium Citrate H.I.E.R, the tissue was incubated with 1 μg/ml of the primary antibody overnight at 4°C. HRP kit was used for detection followed by hematoxylin counterstaining, according to the protocol provided. The image was captured with a 40X objective. Scale bar : 50 μm

ICC staining of purified ab11755 on HeLa cells. The cells were fixed with 4% PFA, permeabilized with a buffer containing 0.1% Triton X-100 and 0.25% BSA, and blocked with 2% normal goat serum and 0.02% BSA. The cells were then incubated with 1 μg/ml of the primary antibody for overnight at 4°C, followed by incubation with 2.5 μg/ml of Alexa Fluor® 594 goat anti-Mouse IgG for one hour at room temperature. Nuclei were counterstained with DAPI, and the slides were mounted with ProLong™ Gold Antifade Mountant. The image was captured with a 40X objective. Scale bar : 50 μm

ab11755 at 1/100 staining mouse gut (small bowel) tissue sections by IHC-P. The tissue was paraformaldehyde fixed and a heat mediated antigen retrieval step in citrate buffer was performed before the tissue was blocked and incubated with the antibody for 45 minutes. An HRP conjugated goat anti-mouse antibody was used as the secondary.

Biological function summary
NEAS plays an important role in maintaining cellular homeostasis. It is known to be part of a larger protein complex which assists in transmitting external signals into internal cellular responses. This protein is essential for balancing cell proliferation and apoptosis contributing to normal tissue maintenance. Its activity regulates several transcription factors that influence gene expression which are vital for cellular growth and differentiation.
Pathways
NEAS is involved in the MAPK/ERK signaling and PI3K-Akt pathways. These pathways are critical for cell growth survival and metabolism. NEAS interacts with key proteins such as Raf and MEK in the MAPK/ERK pathway enhancing signal transduction for cell proliferation. It also shows an association with PDK1 a component of the PI3K-Akt pathway which plays a role in cell survival mechanisms helping cells adapt to environmental stress.
