This antibody stains neural cell adhesion molecule (NCAM) which is present in small cell lung cancer and lung carcinoids. It also reacts with a number of non-small cell lung carcinomas and neuroendocrine and neuronal derived tissues. In addition, it is internalized after binding to its antigen on SCLC cell lines, making it an excellent reagent for tumor imaging in xenograft models. ab9272 reacts with all three isoforms of NCAM.

Immunohistochemistry - Free Floating - Anti-NCAM1 antibody [123C3] (ab9272)

ab9272 at a dilution of 1/1000, staining NCAM (Alexa 488 secondary at 1/2000) on rat brain tissue (30µm thick coronal sections) in free floating IHC (see protocol link for detailed description). Images showing hippocampal neuron body and processes: [A] 20x objective and [B] punctate cytoplasmic staining; 40x objective. No labeling observed following omission of primary antibody.

Sections were viewed using an Axioplan 2 Imaging microscope (Imaging Associates) fitted with 10x, 20x and 40x Plan-Neofluorobjectives (Zeiss, Germany) and images were taken using a AxioCam Hrm digital camera (Zeiss, Germany) and AxioVision software (Imaging Associates).

Flow Cytometry - Anti-NCAM1 antibody [123C3] (ab9272)

Overlay histogram showing SH-SY5Y cells stained with ab9272 (red line). The cells were fixed with 4% paraformaldehyde (10 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab9272, 1µg/1x10⁶ cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x10⁶ cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a positive signal in SH-SY5Y cells fixed with 80% methanol (5 min)/permeabilized with 0.1% PBS-Tween for 20 min used under the same conditions.

Answer

The ability of ab9272 to recognize the nCAM epitope is dependent on the intact conformation of the first fibronectin type III domain (human nCAM aa506-605). The homology of 93.4% between human (http://www.uniprot.org/uniprot/P13591) and hamster (http://www.ncbi.nlm.nih.gov/protein/XP_003498569.1), including high homology between aa506-605 suggests that cross reactivity is likely. We generally like to see 85% or higher homology before we predict that an antibody will recognize its target in a different species, so the 93.4% homology between human and hamster nCAM is promising.

Thank you for your enquiry. Unfortunately, no specific immunogen sequence is known, since the antibody was raised against a membrane preparation of SCLC and not against a specific amino acid sequence.

Question

Thank you for your help. I was particularly interested in products, ab8077, ab9272, ab9018 and possibly ab36 although this last one only reacts with human and not rat. The extracellular domains of NCAM 120, 140 and 180 isoforms are identical but NCAM-120 does not have a cytoplasmic domain as it is linked by a GPI anchor to the membrane. 140 and 180 have cytoplasmic domains of differing lengths. So as long as the antibody recognises something on the extracellular domain it should recognise NCAM-120 as well. Best regards,

Thank you for your enquiry. Ab8077 recognises NCAM-140 and 180 we do not have any information relating to NCAM 120. I am still in the process of sourcing further information regarding Ab9018 and Ab9272 and will be in touch with you again when I have another answer.

Question

I am interested in knocking out the expression of N-CAMs in SH-SY5Y human neuroblastoma cells as part of my studies. Therefore i will be requiring an antibody that i can quantify this knock-down with via western blotting. Do you have an antibody that can detect the three major isoforms-i.e. molecular weights of approx 140, 180 and 220 kDa? Or will i need to purchase two or more antibodies?

Thank you for your enquiry and for your patience. I took a look at the NCAM antibodies that we currently have, and it does not appear that any of them recognize all three major isoforms. For example, ab8077, clone 123C3, recognizes a transmembrane glycoprotein of 140 and 180 kD and ab8596, clone B332 (123A8), reacts with a 135/220 kD glycosylated and sialyated protein. All the information that we have at this time regarding our antibodies is located on the online datasheets. If you have any more questions regarding them, please contact us again.
