Both the ab29486 and ab105784 are giving the same staining pattern which is strange and so I was wandering if you had used a secondary only control on these samples. I am just curious to see if the secondary antibody itself could be giving some of this non-specific background. Also, according to our notes ab28486 will recognize both a 26 & 30kDa band, so I do believe that the intense band signal that is being detected at 26kDa is correct.

Some other protocol variations that might help reduce background are to reduce the primary antibody concentration to 1/500 and also further reduce the total amount of protein loaded into each well. I

If none of the suggestions above prove to be successful, then the we can look into either another replacement or a refund.

As you are not getting the correct signal in western blot using the antibody against rat samples, then I am happy to send you a replacement antibody free of charge.

The only other antibody in our catalogue against Myelin PLP that is at least predicted to work against canine tissue is ab28486:

I have not been able to determine why you are getting a signal at 25kDa. As we had not tested this antibody on canine tissue, we were not sure how it would react, so there is a very good chance that the band is the correct target, just that it is slightly smaller than expected.

I was wondering if you were able to use rat brain lysate as a positive control, since that is what we recommended on the datasheet. If you do not have any rat brain lysate, then please let me know and I will be happy to send some (if you do wish for me to send some, please provide the original Abcam order number).
