      Recombinant fragment (MBP-tag) corresponding to Rat MRP2. This antibody was raised against a fusion protein of the bacterial maltose binding protein and rat MRP2, containing the 202-amino acid C terminal end of the transporter protein.

        All lanes : Anti-MRP2 antibody [M2III-5] (ab15603) at 1/20 dilution

        Lane 1 : Wild-type A549 (Human lung carcinoma cell line) whole cell lysate
        Lane 2 : ABCC2 knockout A549 (Human lung carcinoma cell line) whole cell lysate

        Lysates/proteins at 20 µg per lane.

        Performed under reducing conditions.

        Predicted band size: 185.4 kDa



        Lanes 1 - 2: Merged signal (red and green). Green - ab15603 observed at 210 kDa. Red - loading control, ab52866, observed at 50 kDa.

        ab15603 was shown to recognize ABCC2 in wild-type A549 cells as signal was lost at the expected MW in ABCC2 knockout cells. Additional cross-reactive bands were observed in the wild-type and knockout cells. Wild-type and ABCC2 knockout samples were subjected to SDS-PAGE. The membrane was blocked with 3% Milk. Ab15603 and ab52866 (Rabbit anti-alpha Tubulin loading control) were incubated overnight at 4°C at 1/20 dilution and 1/20000 dilution respectively. Blots were developed with Goat anti-Mouse IgG H&L (IRDye® 800CW) preabsorbed ab216772 and Goat anti-Rabbit IgG H&L (IRDye® 680RD) preabsorbed ab216777 secondary antibodies at 1/20000 dilution for 1 hour at room temperature before imaging.

      Question

      I have a question concerning the antibody ab15603 [M2III-5] MRP-2.

      I have seen that some publications used an antibody from other companies like Enzo identified M2III-5. It was used to detect the MRP-2 protein in mouse. On the Enzo website, it is indicated that this clone react with human, rat and mouse. However, on your website, the same clone number is indicated to not react with mouse.

      I want to know if the clone numbers (ex: M2III-5) are in fact representing the exact same thing between every companies?

      I hope that my question is clear.

      The note regarding mouse specificity is based on data from the following reference, which includes a description of the clone's development and characterization:

      Table 1 has a summary of species reactivity for this clone and several others.

      Further characterization may have shown reactivity with other murine samples, in other applications, and this may account for the information on the datasheets of other vendors, but we do not have this data.

      It is possible that there is more than one clone raised against MRP2 designated "M2III-5" but this seems less likely than the possibility of differences in testing conditions and samples across different laboratories.

      Thank you very much for your inquiry and your patience while waiting for our answer.

      I have inquired with the laboratories of the polyclonal antibodies, whether the immunogen sequence is conserved with the pig sequence. This would then provide a good basis for testing the antibody.

      The ab110740 has unfortunately only 79%homology with the pigsequence and we can not recommend it therefore.

      Also for the ab58709, the alignment is only 62%, and we can not recommend it neither. I am very sorry.

      For the monoclonal antibodies, it is more difficult to predict, as the epitopes have not been mapped. As however the overall sequence alignment is relatively good, they might have also good changes to crossreact with the pig protein.       The ab3373 and the ab15603 have been raised against the human protein sequence 1339-1541. There are many conserved residues.
      The ab24106 and the ab130385 have been raised against the human sequence 860-950, which seems less conserved than the C terminal. The ab3372 has been raised against human sequence 215 to 310, again, a less conserved region than the C terminus.

      So in summary, I would think either the ab3373 or ab15603 most likely to crossreact with the pig protein from the choice of antibodies we have. This has however not been tested, and it can well be that they will not react with the pig protein.

      Question

      I have used your M2III-5 antibody to examine the expression of mouse Mrp2 protein. Your product info sheet stated that M2III-5 can cross-react with rat, mouse and human Mrp2. Strangely, on our western blot a 170KD band was shown in both the wild type and the Mrp2 knockout samples. Honestly, I would think that band is the Mrp2 band if I did not have the Mrp2 KO sample as a negative control. We have work with M2III-6 a lot in our research, M2III-6 work beautifully in the WT and Mrp2-/- rats. From literature, Scheffer GL et al developed the clone for M2III-5 antibody and in their paper "M2III-5 was shown NO cross-reactivity to the mouse Mrp2" (table 1 of the paper, Cancer Research 2000, 60:5269-77). Also from the paper, M2III-5 and M2III-6 recognize the same aa sequence (1336-1541). M2III-6 has no cross-reactivity with mouse Mrp2 either although both M2III-5 and 6 can react with rat Mrp2 very well. I wonder if there is any difference between your M2III-5 antibody and the authors'. From my conversation with one of your tech support person this morning, you use the same clone as the authors. I personally think they should have the same reactivity. Could you please explain me why your antibody has different reactivity from author's? Is there any possibility that your M2III-5 can recognize a band other than Mrp2 in the mouse sample? Have you confirmed the specificity of your M2III-5 in mouse samples? P.S. attachment of M2III-5 results with moue liver sample.
