MED12L also known as Mediator of RNA polymerase II transcription subunit 12-like is a protein encoded by the MED12L gene in humans. It has a molecular weight of approximately 250 kDa. MED12L expression occurs widely throughout the body with a notable presence in tissues such as the brain liver and kidney. This protein plays a mechanical role in the transcriptional regulation interacting with multiple components during the regulatory process.
Biological function summary

The MED12L protein functions as part of the Mediator complex which is essential for the transcription of nearly all RNA polymerase II-dependent genes. The Mediator complex acts as a bridge between gene-specific regulatory proteins and the polymerase machinery facilitating gene expression. Through this complex MED12L contributes significantly to regulating developmental processes and cellular differentiation.

Pathways

MED12L is intimately involved in the Notch signaling pathway and the Wnt signaling pathway. These pathways are critical for cell fate determination proliferation and communication during embryonic development and in adult tissue homeostasis. MED12L interacts with other Mediator complex proteins such as MED1 and MED19 to facilitate these pathways and ensure proper signal transduction and transcriptional regulation.

MED12L has associations with intellectual disabilities and certain cancers. Mutations or dysregulation of MED12L can lead to disrupted Mediator complex function impacting cognitive development and potentially contributing to tumorigenesis. In cancer MED12L-related pathways may intersect with pathways involving proteins such as β-Catenin and Notch receptors therefore playing a role in oncogenic transformation and disease progression.
