Immunofluorescent analysis of HEK293 cells transfected with pFin-EF1-mCherry vector labeling mCherry using ab205402 at 1/1000 dilution (green). Red : mCherry; Blue : blue DNA stain. Note : Most HEK293 cells are not transfected so only the nucleus of these cells can be visualized with a blue DNA stain. Cells which are transfected with mCherry are bright red. Green antibody staining is only seen cells which express Cherry, as expected, and the superimposition of the green and red results in an orange signal.

Immunocytochemistry/ Immunofluorescence - Anti-mCherry antibody (AB205402)

Immunocytochemistry/ Immunofluorescence analysis of Human ES cells with mCherry gene reporter labeling mCherry with ab205402 at 1/1000 dilution. Cells were fixed with paraformaldehyde and permeabilized with 0.1% Triton-X. Blocking was carried out with 10%BSA for 1 hour at 4°C, followed by incubation with ab205402 at 1/1000 for 12 hours at 4°C. A polyclonal goat anti-chicken Alexa Fluor^® 488 was used as the secondary antibody at 1/100 diluton.

Western blot - Anti-mCherry antibody (AB205402)

Western blot analysis of HEK293 cell lysates using chicken pAb to mCherry, CPCA-mCherry, dilution 1 : 2,000, in green, and rabbit pAb to GAPDH, RPCA- GAPDH, dilution 1 : 5,000, in red. The CPCA-mCherry antibody recognizes tdTomato and mCherry proteins revealing major bands at about 60kDa (tdTomato) and 30kDa (mCherry), but does not recognize GFP. The red band at 37kDa corresponds to GAPDH protein detected using a mouse monoclonal antibody used here as a gel loading control.

The WB image was produced on the LiCor Odyssey imaging system. Secondary antibody conjugates are reconstituted to 1mg/mL and are diluted to working concentrations from this stock solution.
