JMJD1C also known as KDM3C is a member of the Jumonji domain-containing proteins family. It acts as a histone demethylase specifically targeting histone H3 at lysine 9 (H3K9). This enzymatic function plays a role in epigenetic regulation by removing methyl groups from histone proteins influencing chromatin structure and gene expression. The protein has an approximate molecular mass of 170 kDa. It is expressed in various tissues with significant levels found in the testis brain and other central nervous system components.
Biological function summary

JMJD1C regulates gene expression linked to spermatogenesis metabolic processes and neurodevelopmental signaling. It interacts with components of transcriptional complexes having a role in the demethylation of histones to facilitate transcription. The protein also controls hormonal responses by influencing androgen receptor signaling pathways which is important in reproductive tissue function and development.

Pathways

JMJD1C integrates into metabolic and developmental regulatory networks. It is actively involved in the androgen receptor signaling pathway and the Wnt signaling pathway. Its interactions with proteins such as AR (androgen receptor) and components of the Wnt signaling orchestrate gene transcription impacting cellular growth and differentiation. By modulating these pathways JMJD1C links epigenetic modifications to broader physiological processes.

JMJD1C has been associated with cancer and neuropsychiatric disorders. Aberrant expression or mutation of JMJD1C can contribute to prostate cancer given its role in androgen receptor pathways and it has also been implicated in schizophrenia where it may influence neuronal gene expression. The protein's interaction with the androgen receptor and other transcription factors highlights its significance in these pathologies suggesting potential as a therapeutic target.
