I am trying to use this antibody in a Western blot and I am having some difficulty in determining if the antibody is recognizing insulin. Can you tell me how the ELISA testing was conducted, for example? Did you use both recombinant proinsulin and insulin?

Thank you for your enquiry. This antibody was tested along with ab8304 and ab28063 in sandwich ELISAs against recombinant proinsulin and insulin. ab8305 and ab8304 are directed to different antigenic determinants. The Kd for ab8304 is 8,1 x 10-8M. The Kd for ab8305 is 6,3 x 10-8M. The antibodies were used for detection of rat/mouse insulin/proinsulin in two-site enzyme immunoassays and IHC. We recommend to use ab8304 for coating and ab8305 for labeling. The antibodies have not been tested in Western blot. Thank you for your enquiry. The antibody reacts with whole molecule of mouse insulin and proinsulin. The antibody is working in immunohistochemistry on frozen sections. As immunogen we have used native human insulin.
