This data was developed using ab300081, the same antibody clone in a different buffer formulation.
ING3 was immunoprecipitated from 0.35 mg HeLa (human cervix adenocarcinoma epithelial cell) whole cell lysate 10 μg with ab300081 at 1/30 dilution (2μg in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab300081 at 1/1000 dilution. VeriBlot for IP secondary antibody(HRP)(ab131366) was used at 1/5000 dilution.
Lane 1 : HeLa (human cervix adenocarcinoma epithelial cell) whole cell lysate 10 μg
Lane 2 : ab300081 in HeLa whole cell lysate
Lane 3 : Rabbit monoclonal IgG (ab172730) instead of ab300081 in HeLa whole cell lysate
Blocking and dilution buffer and concentration : 5% NFDM/TBST.
Exposure time : 24 seconds
Lysates should be made freshly and used in IP immediately to minimize protein degradation.

Biological function summary
ING3 influences gene transcription and chromatin remodeling as a part of the NuA4 histone acetyltransferase complex. This complex is involved in the acetylation of histone H4 and H2A facilitating an open chromatin structure that permits transcriptional activation. ING3 is important in the regulation of cell cycle progression and apoptosis. It modulates p53 and other key cellular pathways underpinning its role in maintaining normal cellular homeostasis and preventing cancerous transformations.
Pathways
ING3's function supports significant biological processes like DNA repair and apoptosis. It is part of the p53 signaling pathway which plays a pivotal role in the cellular response to DNA damage. ING3 acts alongside proteins such as p21 and MDM2 in the p53 pathway. Furthermore ING3 interfaces with the MAPK/ERK signaling pathway contributing to its regulation of cell division and survival. Through these pathways ING3 helps orchestrate a balanced response to cellular stress and damage.
