Stable inducible HepG2-IFN-λ4-ISRE-Luc cells were induced with 0.5 mg/ml doxycycline for 16hours and then treated with PBS (negative control) and rabbit IgG (20μg/ml) for 22 hours and assayed for luciferase expression. α-IFN-λ4 (ab196984) blocks the ability of IFN-λ4 to induce of ISRE-Luc reporter by 70%.

IFN-λ4 overexpression induces ISG expression. (A,B) Huh-7 cells were transfected with control or IFN-λ4-expressing plasmids. After 72 hours, the cell lysates (A) and culture supernatants (B) were harvested, and protein expression were analysed by immunoblot. For the culture supernatants, immunoprecipitation with anti-FLAG-conjugated beads were used to pull down the FLAG-tagged IFN-λ4 protein. The data are representative of two independent experiments. (C,D) Huh-7 cells were transfected with control, IFN-λ1-, IFN-λ4-, or IFN-β-expressing plasmids. After 96 hours, the cells were harvested, and protein expression or gene expression were analysed by immunoblot (C) or real-time qPCR (D). (E) Huh-7 cells were transfected with IFN-λ4-expressing plasmids. The cells were harvested at the indicated time points, and gene expression was analysed by real-time qPCR. (F) Huh-7 cells were infected with JFH1 HCVcc at 0.5 MOI followed by transfection with control or IFN-λ4-expressing plasmids. After 72 hours, the cells were harvested, and HCV RNA titer was analysed by real-time qPCR. (G,H) Huh-7 cells were transfected with control, IFN-λ4-, or IFN-λ4 TT-encoding plasmids. IFNL4 also known as Interferon Lambda 4 plays an important role in the immune system. This protein with a mass of approximately 22 kDa is part of the type III interferon family. It expresses mainly in liver cells and epithelial tissues. IFNL4 is involved in the antiviral defense mechanism of the body. It shows activity by binding to the IFN lambda receptor complex initiating a response that helps control viral infections.
Biological function summary

IFNL4 acts as a cytokine and contributes to modulating immune responses. It is not part of a complex but interacts closely with the IFN lambda receptor to activate Janus kinase and signal transducer and activator of transcription (JAK-STAT) signaling pathways. This activation leads to the promotion of antiviral responses in infected cells and limits viral spread. Understanding its signaling function clarifies its important impact on immune defense.

Pathways

IFNL4 is fundamental to the JAK-STAT signaling pathway and to the innate immune response pathway. These pathways mediate cellular responses to viral infections. IFNL4 interacts with other proteins like IFNLR1 IL10RB and STAT1 which are essential for these pathways. By engaging these pathways IFNL4 helps control the expression of genes involved in antiviral defense such as those coding for interferon-stimulated genes (ISGs).

IFNL4 has strong connections to hepatitis C virus (HCV) infection and liver fibrosis. Researchers found that the expression of IFNL4 can influence the outcome of HCV infection affecting viral clearance and treatment response. Another related protein IFNL3 also interacts with these pathways and together they provide insights into the genetic factors influencing liver fibrosis progression. Understanding IFNL4's role provides opportunities to develop targeted therapies for these conditions.
