    In cooperation with other chaperones, Hsp70s stabilize preexistent proteins against aggregation and mediate the folding of newly translated polypeptides in the cytosol as well as within organelles. These chaperones participate in all these processes through their ability to recognize nonnative conformations of other proteins. They bind extended peptide segments with a net hydrophobic character exposed by polypeptides during translation and membrane translocation, or following stress-induced damage.

Question

The datasheet for ab108416 claims that the target of the antibody is HSPA2. To our knowledge, expression of this protein is present only in testes, but the datasheet shows positive IHC signal in colon tissue, where it should not be. Is the data mis-placed, or was the target of the antibody incorrectly specified?

Answer

Thank you for contacting us about antibody ab108416. The ab108416 datasheet does list, as an alternative name for the target of the antibody, "Testis-specific heat shock protein-related antibody". However, the colon data is not misplaced. The antibody was raised against a fragment of human HSPA2 and was in fact validated for IHC on normal human colon tissue.

I found two recent papers that challenge testes-specific expression, at least for human samples.

From the abstract: "HSPA2 is a human counterpart of the testis-specific rodent Hst70/Hsp70.2 gene. In contrast to the latter, the expression of the human HSPA2 gene is not limited to the testis...".

The same lab published the following tissue microarray study, using their own rabbit polyclonal antibody:

On the other hand, characterization of ab108416 has not been carried out on knockout tissue or on siRNA-treated cells, so cross-reactivity with, for instance, HSPA1 is possible. The western blot data for various lysates shows detection of a single band, but HSPA2 and HSPA1 are the same size.
