Western blot - HRP Anti-Peroxiredoxin 2/PRP antibody [EPR5154] (AB197042)
This blot was produced using a 4-12% Bis-tris gel under the MES buffer system. The gel was run at 200V for 35 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 3% milk before being incubated with ab197042 overnight at 4°C. Antibody binding was visualised using ECL development solution ab133406.

HEK-293 (Human) Whole Cell Lysate (ab52256) at 10 µg

Lane 3:
LNCaP (Human prostate adenocarcinoma cell line) Whole Cell Lysate - clone FGC at 10 µg

SHSY-5Y (Human neuroblastoma cell line) Whole Cell Lysate at 10 µg

Western blot - HRP Anti-Peroxiredoxin 2/PRP antibody [EPR5154] (AB197042)
ab197042 was shown to specifically react with PRDX2 in wild-type HAP1 cells as signal was lost in PRDX2 (Peroxiredoxin 2) knockout cells. Wild-type and PRDX2 (Peroxiredoxin 2) knockout samples were subjected to SDS-PAGE. ab197042 and ab184095 (Mouse monoclonal [mAbcam 9484] to GAPDH - Loading Control (Alexa Fluor® 680) loading control) were incubated overnight at 4°C at 1/3000 dilution and 1/1000 dilution respectively. The loading control was imaged using the Licor Odyssey CLx prior to blots being developed with ECL technique.

Biological function summary
Peroxiredoxins like PRDX2 function by breaking down hydrogen peroxide and organic hydroperoxides safeguarding cells from oxidative harm. PRDX2 often forms homodimers or higher-order oligomers which influence its catalytic efficiency and chaperone activity. Within the cell it not only acts independently but also associates with other cellular components reflecting its participation in important cellular processes including cell proliferation and apoptosis.
