补充信息

This supplementary information is collated from multiple sources and compiled automatically.

HIP1 also known as Huntingtin-interacting protein 1 is a protein with a mass of approximately 116 kDa. HIP1 is an adapter protein involved in the endocytosis process. It interacts with clathrin and AP2 adaptor complex assisting in the invagination and fission of clathrin-coated pits. HIP1 is present throughout various tissues including the brain and immune cells indicating its widespread role in cellular mechanisms.
Biological function summary

HIP1 plays an essential role in the modulation of receptor-mediated endocytosis. It is part of a larger complex with epsin and other proteins facilitating the internalization of extracellular signals. The protein's interaction with actin and lipid membranes suggests its function in reorganizing cytoskeleton arrangements vital for cell movement and shape changes. Cellular homeostasis and signal transduction rely on HIP1's ability to mediate these activities.

Pathways

HIP1 integrates into the PI3K/AKT signaling impacting cell survival and apoptosis. Its activity in clathrin-mediated endocytosis also links it to the receptor tyrosine kinase (RTK) pathway. In these pathways HIP1 assists in the regulation of growth factor receptors collaborating with proteins such as GRB2 and SHC1. These interactions ensure proper cellular responses to external stimuli.

HIP1 has associations with cancer and neurodegenerative diseases. In cancer overexpression of HIP1 enhances cell proliferation linked to its role in receptor trafficking and signaling. HIP1's interaction with huntingtin the protein mutated in Huntington's disease suggests its involvement in neurodegeneration. Such connections emphasize the protein's potential as a target for therapeutic intervention highlighting HIP1's impact on pathological conditions.

靶点信息

Plays a role in clathrin-mediated endocytosis and trafficking (PubMed : 11532990, PubMed : 11577110, PubMed : 11889126). Involved in regulating AMPA receptor trafficking in the central nervous system in an NMDA-dependent manner (By similarity). Regulates presynaptic nerve terminal activity (By similarity). Enhances androgen receptor (AR)-mediated transcription (PubMed : 16027218). May act as a proapoptotic protein that induces cell death by acting through the intrinsic apoptosis pathway (PubMed : 11007801). Binds 3-phosphoinositides (via ENTH domain) (PubMed : 14732715). May act through the ENTH domain to promote cell survival by stabilizing receptor tyrosine kinases following ligand-induced endocytosis (PubMed : 14732715). May play a functional role in the cell filament networks (PubMed : 18790740). May be required for differentiation, proliferation, and/or survival of somatic and germline progenitors (PubMed : 11007801, PubMed : 12163454).
