    ICC/IF image of ab53940 stained HepG2 cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal donkey serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab53940, 5µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 donkey anti-goat IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

Thank you for taking the time to complete our questionnaire and contact us. I am sorry to hear you have had difficulty obtaining satisfactory results from this antibody.
The details you have kindly provided will enable us to investigate this case for you and this is also helpful in our records for monitoring of quality.
Reviewing this case, I would like to offer some suggestions to help optimise the results from ab53940. I would also appreciate if you can confirm some further details:
1) The lysis buffer seems unusual (we usually recommend RIPA buffer), but if it works for you it is fine. However, we strongly recommend adding freshly prepared protease inhibitors. Degradation of proteins can occur very quickly, leading to an unspecific banding pattern which might explain the results you observe. For details see our WB guide attached.
2) We usually recommend 20-30 ug per lane, so the 30 ug you loaded should be absolutely fine to avoid overloading which might increase unspecific signals as well.
3) When testing our antibodies, our lab uses 5% BSA as a blocking reagent, so I recommend switching to this instead of milk. For unknown reasons some antibodies give stronger, more specific signals on blots blocked with BSA instead of milk, so doing this may improve the results you are seeing, and reduce the non-specific bands. An example of the above is the western blot obtained with the Abcam GAPDH antibody ab9385 : https://www.abcam.com/gapdh-antibody-hrp-loading-control-ab9385.html#GAPDH-Primary-antibodies-ab9385-3.jpg (or use the following: www.abcam.com/ab9385). Also, it is then recommended to use the same blocking reagent also in the antibody dilution buffers (i.e. no mixing of milk/BSA).
4) Have you performed a no primary control to see if the additional bands are caused by unspecific binding of the secondary antibody?

i purchased the GSTA-1 antibody from your company. recently, i run my western blot and i get a specific band at˜20-26kda. but at the same time i get multiple non specific bands which are more intense compared to the specific bands. i tried with increase with either the protein amount or antibody concentration, but yet it doesnt improve the result.can i noe is there any solution to solve this problem?? or any of your customer encounter this problem before and how they solve it? thank you for your help...thank you and have a nice day!
