    Inquiry: Dear Sir/ Madam, Sorry to disturb you. Our customer raised an enquiry about ab9741. She already used ab9741 to conduct the ICC/IF assay with primary culture cell--Rat Cerebral Cortex tissue. She thinks the expression of GM-CSF protein should in the nuclear and cytosol of her sample. But the result shows the specific signal just in the nuclear. Besides, she also wants to know is there any customer conducted the same experiment just like her or not? If yes, could you please offer any suggestions to improve her results? Furthermore, could you please offer any selections which can be a positive control in her experiment?     What is the difference between neutralizing and inhibition functions stated in Abcams datasheets.
    The datasheet also states the required antibody concentration for a 50% inhibition of bioactivity, wouldan increase inconcentration lead to a linear increase in inhibition?

    The neutralizing and inhibition functions stated on the datasheet of the antibodies ab9741 andab64715 relate to the assays which have been performed to validate their activities.

    With the Anti-GM-CSF antibody (ab9741), the antibody has been used in a neutralising assay as described in the protocol attached to this email.

    Whilst with the Anti-TGF beta 1 antibody [2Ar2] (ab64715) has been used in an inhibition of proliferation assay. This was carried out asdescribed in Tsang, M. et al., (1995) Cytokine 7:389.The dose response curves are given in figures 1 and 2 of the attached document.The response is linear over a small range.

    Thank you very much for your inquiry.

    The immunogen of ab9741 was the whole GM-CSFprotein of mouse. Mouse GM-CSF only shows 71% homology with rat as a whole protein and therefore I cannot recommend to use this antibody for rat samples.

    We do have two antibodies that have a immunogen (peptide) that is taken from a part of the GM-CSF human or ratsequence that shows a higher homology to the rat sequence. Both antibodies are predicted to react with rat and I would be happy to issue a discount code for either of them.

    Maybe your customer would like to review the datasheets and choose one of them for testing in rat?

    The GM-CSF protein is expected to run around 16 kDa, however there are 4 known glycosylation sites, each adding around 2 kDa to the protein, so the protein generally runs around 24 kDa on a Western blot.

    Unfortunately, we only test against the recombinant murine GM-CSF and do not test tissue samples. In the naturally occurring GM-CSF there are potential glycolisation sites which could cause the protein to migrate higher on a WB. Sorry we could not be of more assistance on this one.
