In particular it would be helpful to receive further information on the purified protein that you are using. Is this a recombinant protein, was this full length protein? Or is it purified endogenous native protein? Has it been reduced and denatured?

It is possible that this antibody is not quite as sensitive as the other one you are using and you may need to load more protein for detection.

For the blot with which you used ab8027, could you confirm the quality of the sampleand transfer to the membrane from that lane was checked using commassie on ponceu red staining?
