Immunocytochemistry/ Immunofluorescence - Anti-GCDFP 15 antibody [EP1582Y] (ab62363)

Immunocytochemistry/Immunofluorescence analysis of MCF-7 (human breast carcinoma) cells labelling GCDFP 15 with ab62363 at 1/100. Cells were fixed with 4% Paraformaldehyde and permeabilized with 0.1% Triton-X. ab150077, Alexa Fluor^® 488-conjugated goat anti-rabbit IgG (1/1000) was used as the secondary antibody. Cells were counter-stained with ab7291 Anti-Tubulin antibody (1/1000) followed by ab150120 AlexaFluor^®594 Goat anti-Mouse secondary (1/1000). Nuclei were counterstained with DAPI (blue).

For negative control 1, ab62363 at dilution of 1/100 was used followed by anti-mouse secondary antibody (ab150120) at 1/1000 dilution. For negative control 2, ab7291; 1/1000 was used followed by anti-rabbit secondary antibody (ab150077) at 1/1000 dilution.

Intracellular Flow Cytometry analysis of T47D (human mammary gland ductal carcinoma) (pink) with ab62363 at dilution of 1/20. The secondary antibody used was goat anti rabbit IgG (FITC) at dilution of 1/150. Cells were fixed with 2% paraformaldehyde. Isotype control antibody used was Rabbit monoclonal IgG (green).

Question

Hi, I would like to know if the antigen "GCDFP 15 peptide (ab48069)" can be linked by the antibodies "ab62363". I'm actually trying to perform a direct ELISA using the previous antibody, since we found it to perfectly detect PIP in our human saliva samples by using wester blot technique. My first attempt to use this antibody for the ELISA was good, so now I only need a PIP (antigen) to be used as a standard to perform serial dilution and to obtain a standard curve.

Answer

The antibody ab62363 has an immunogen near the N-terminus of human GCDFP 15. The peptide ab48069 you mention however is derived from the internal region of GCDFP 15 and therefore ab62363 will not bind to it.

I have been in contact with the lab and they have agreed to add a new product to the catalogue, another GCDFP 15 peptide. This GCDFP 15 peptide is the blocking peptide of ab62363 and therefore ab62363 should bind to it since the peptide was used to raise this antibody. The catalogue number of this peptide is ab155648 and costs €180.
