    • 功能

      Receptor for Wnt proteins. Most of frizzled receptors are coupled to the beta-catenin canonical signaling pathway, which leads to the activation of disheveled proteins, inhibition of GSK-3 kinase, nuclear accumulation of beta-catenin and activation of Wnt target genes. A second signaling pathway involving PKC and calcium fluxes has been seen for some family members, but it is not yet clear if it represents a distinct pathway or if it can be integrated in the canonical pathway, as PKC seems to be required for Wnt-mediated inactivation of GSK-3 kinase. Both pathways seem to involve interactions with G-proteins. May be involved in transduction and intercellular transmission of polarity information during tissue morphogenesis and/or in differentiated tissues. Interacts specifically with Wnt5A to induce the beta-catenin pathway.

    Thank you for your reply.The cells I am using are rat intestinal epithelial cells. They are lysed in RIPA (although I have tested a number of lysis buffers but with no diffrerence between them). They are either sonicated or dounced (again no difference in bands seen) and the samples are run on standard SDS-PAGE gels in reducing conditions. The block is 5% BSA followed by 5% BSA with 1:200 Fz5. The secondary is 4% milk. The lot number associated with the new vials (the old ones were discarded) is GR64385-1.

    I have used up quite a lot of the antibody by WB (but also now by IF) to try and detect a signal which has been highly frustrating and I wish I had some of the previous antibody left to directly confirm that its not working, but I do not have this. A replacement (of a different lot number) would be highly appreciated if possible please.

    Dear Abcam,

    I have recently purchased 2 vials of Frizzled 5 antibody (ab14475).

    You say that your predicted size is 63kDa yet your blot shows 55kDa and +200kDa bands. Please could you enlighten me on the identity of these bands as I seem to be only getting a doublet at 250kDa (although with the previous batch of this antibody I was also able to see the 55kDa band).

    Thank you very much for contacting us with your question, and for your patience while I have been in touch with the lab regarding this antibody.

    The predicted molecular weight is based only on the precursor amino acid sequence of Frizzled 5, and after the signal peptide is cleaved the protein will have a lower molecular weight. The lower band below 62 kDa corresponds to the cleaved mature protein. The lab scientists have not further characterized the higher band above 200 kDa, and ithas been hypothesized that this isan aggregate of the protein, but I haven't found support for this in the literature. The image shows a transfected lysate, so perhaps it is a product of the transfection, but it may also be a non-specific band. Could you tell me what kind of lysates you are using? How are the samples prepared? What blocking solution is used, and what dilution of antibody has been tried? What is the lot number associated to each of these vials that you've tried?
