Western blot - Anti-FKBP15 antibody (AB14432)
Antibodies : ab14432 used at 0.2 mcg/ml for WB in A and B and 2 mcg/mg extract for IP (B), ab14433 and ab14434 used at 2 mcg/mg extract for IP. Detection : Chemiluminescence with an exposure time of 25 minutes (A) and 10 minutes (B).
In figure B the Immunoprecipitation was done using ab14432 (Lane 3), ab14433 (Lane 4) and ab14434 (Lane 5) at 2μg/mg of extract.

Biological function summary
FK506-binding protein 15 assists in modulating protein-protein interactions due to its isomerase activity. FKBP15 is involved in the secretory pathway and may associate with various intracellular complexes to exert its function. This association enhances its ability to regulate proteins trafficking and processing within the endoplasmic reticulum ensuring proteins reach their correct destinations within the cell.
Pathways
FKBP15 plays a role in intracellular signaling and immune regulation. It participates in the calcineurin signaling pathway where it regulates calcium ion-dependent processes linking it to proteins such as calcineurin and NF-AT. FKBP15 also engages with the endoplasmic reticulum-associated degradation (ERAD) pathway contributing to the degradation of misfolded proteins and maintaining cellular homeostasis.
