I could replace the antibody at no cost however as we do not have received complaints about this lot so I am wondering the problem might be either with the expression of luciferase or with the dilution of the antibody. The 1/5000 dilution recommended on the datasheet is for guidelines only so 1/2000 dilution or loading high concentration of protein might be tried for optimization.

The provided data does not show which cell type was used for lysates preparation so I would like to ask if you have confirmed the expression of luciferase by other methods. if yes, could you share the details?
