Substrate recognition component of a SCF (SKP1-CUL1-F-box protein) E3 ubiquitin-protein ligase complex which mediates the ubiquitination and subsequent proteasomal degradation of target proteins. Probably recognizes and binds to phosphorylated target proteins. Involved in the degradation of cyclin-E, MYC, NOTCH1 released notch intracellular domain (NICD), and probably PSEN1. -

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of rat brain tissue labeled with AB317368 at a 1:200 dilution followed by conjugation to the secondary antibody and DAB staining.

Immunocytochemistry/ Immunofluorescence - Anti-Fbxw7 Antibody (ab317368)Immunocytochemistry/ Immunofluorescence analysis of 4% Paraformaldehyde-fixed, Triton X-100 permeabilized HepG2 cells staining Fbxw7 using AB317368 at a 1:100 dilution, for 90 minutes at 37°C, followed by a conjugated Goat Anti-Rabbit IgG antibody at 37°C for 90 minutes. DAPI (blue) was used to stain the cell nuclei.
Blocking buffer (normal goat serum) at 37°C for 20 min. -
Flow Cytometry (Intracellular) - Anti-Fbxw7 Antibody (ab317368)Mouse spleen cells were fixed with 4% PFA for 10min at room temperature, permeabilized with ice-cold methanol for 20 min at room temperature, and incubated in 5% BSA blocking buffer for 30 min at room temperature. Cells were then stained with AB317368 at 1:50 dilution in blocking buffer and incubated for 30 min at room temperature, washed twice with 2% BSA in PBS, followed by secondary antibody incubation for 40 min at room temperature.
Acquisitions of 20,000 events were performed.
Cells stained with primary antibody (green), and isotype control (orange).
