    Factor XIII is activated by thrombin and calcium ion to a transglutaminase that catalyzes the formation of gamma-glutamyl-epsilon-lysine cross-links between fibrin chains, thus stabilizing the fibrin clot. Also cross-link alpha-2-plasmin inhibitor, or fibronectin, to the alpha chains of fibrin.

      Overlay histogram showing A549 cells stained with ab16956 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab16956, 1µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed.

    Question

    I can’t see an attachment with the questionnaire but I have attached my protocol.

    The issue that I am having is the AC-1A1 antibody (cat no. Ab-1834) does not detect the factor XIII-A subunit in normal platelet free plasma. The antibody has worked in this experiment because it has bound to factor XIII in Fibrogammin.

    My worry is that the antibody is unable to detect factor XIII-A in plasma due to interactions with other proteins and I was wondering whether you would be able to confirm whether this is the case or not? I have been unable to find literature where this antibody has been used to detect FXIII-A in plasma by Western Blot.

    All calculations and reagents are stated in the protocol.

    Thank you for your phone call and email.

    I apologize as I forget to attach the questionnaire. I have looked into the details and would like to make following suggestions;

    ab1834 was able to detect band in Fibrogammin samples so I assume the antibody is fine. The problem might with the plasma samples or the protocol. We have not tested this antibody with plasma sample so we may not be able to recommend you specific protocol.

    Factor X111 is present in plasma as well as in platelets. Please use normal plasma along with plasma without the platelets (sample of your interest). Placental tissue lysates can also be used as positive control.

    Try increasing the antibody concentration.

    In the following publication (http://ard.bmj.com/content/53/4/256.full.pdf+html) researcher have tried non-reduced conditions with plasma samples, you can try this.

    Check the protein transfer using ponceau method.

    This antibody was used in the following publication:

    Ulici V et al. Regulation of gene expression by PI3K in mouse growth plate chondrocytes. PLoS One 5:e8866 (2010). WB, IHC-P; Mouse. (PubMed: 20111593)



    They used this protocol with ab1834 with success:

    3% H2O2 for 15 min at room temperature, followed by incubation in preheated (2 min at 100°C) 10 mM sodium citrate solution (pH 6.0) for 30 min at 97°C. They were then blocked with 5% serum. Sections were incubated with 0.10 mg/ml factor XIIIa primary antibody over night at 4°C.

Further to our liveperson discussion yesterday morning I have performed some literature searching and in the following publication factor XIIIA was studied using BOTH reducing and non-reducing conditions. Whilst it is chicken factor XIIIA I consider it your best option that you electrophorese this protein under both conditions to determine homo- and hetero-dimer formation as they have in the publication: Chicken coagulation factor XIIIA is produced by the theca externa and stabilizes the ovarian follicular wall.     Question

    I have a customer's enquiry regarding ab1834. She wants to used this antibody for IHC-Paraffin, she would like to know what is the best antigen retrieval method, eg enzymatic retrieval. I told her about the method using chymotrypsin, however she was wondering if she could use protease k instead as its much quicker. I am not familiar with using protease k, could you suggest a protocol that I can forward onto her?

    Thank you for your enquiry regarding ab1834 and your interest in our products. This particular antibody to Factor XIIIa does not require any antigen retrieval for paraffin sections. Proteinase K is more suitable for in situ hybridization and not for IHC. Generally for IHC, the safest enzymatic treatment is Trypsin or Pepsin.

    I am carrying out research on fibrous lesions of the oral mucosa and am interested in using ab1834 (anti Factor XIIIa) on formalin fixed paraffin-enbedded tissue. Since these are very fibrous lesions, I have in the past experienced problems with loss of tissue architecture when using heat mediated antigen retrieval. I would therefore like to use enzymatic antigen retrieval. Is this recommended for this antibody?
