Immunocytochemistry/ Immunofluorescence - Anti-Epac1 antibody (AB21236)

ICC/IF image of ab21236 stained MCF7 cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab21236, 5µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

Epac1 also known as Exchange Protein directly Activated by cAMP 1 plays an important role in cellular signaling. It is a guanine nucleotide exchange factor with a molecular mass of about 105 kDa. This protein activates Rap1 and Rap2 small GTPases by promoting the exchange of GDP for GTP. Epac1 is widely expressed in various tissues including the heart brain and immune cells. Its distribution indicates its involvement in multiple physiological processes.

Through simultaneous binding of PDE3B to RAPGEF3 and PIK3R6 is assembled in a signaling complex in which it activates the PI3K gamma complex and which is involved in angiogenesis. Plays a role in the modulation of the cAMP-induced dynamic control of endothelial barrier function through a pathway that is independent on Rho-mediated signaling.
