Involved in cytoskeletal rearrangements required for phagocytosis of apoptotic cells and cell motility. Acts in assocation with DOCK1 and CRK. Was initially proposed to be required in complex with DOCK1 to activate Rac Rho small GTPases. May enhance the guanine nucleotide exchange factor (GEF) activity of DOCK1. -

Lane 1 : Human Frontal Cortex cell lysate (35µg protein in RIPA buffer).
Lane 2 : Mouse cell lysate (35µg protein in RIPA buffer).
Lane 3 : Rat cell lysate (35µg protein in RIPA buffer).Primary incubation was 1 hour. Detected by chemiluminescence.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-ELMO1 antibody (ab2239)IHC image of ab2239 staining in human normal lymphoid formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol B. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab2239, 10µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times. -
