EDEM1 also known as ER degradation-enhancing alpha-mannosidase-like protein 1 is a protein that plays a role in the endoplasmic reticulum-associated degradation (ERAD) pathway. Its molecular mass is approximately 98 kDa. EDEM1 is expressed in various tissues but is highly present in the liver and pancreas. This protein helps to identify and target misfolded glycoproteins for degradation ensuring quality control within the endoplasmic reticulum.
Biological function summary

EDEM1 accelerates the removal of incorrectly folded proteins by promoting their retrotranslocation and subsequent degradation by the proteasome. It is not part of a stable complex but coordinates with ER chaperones to enhance the dislocation of substrates from the ER to the cytosol. EDEM1 regulates glycoprotein quality control helping to maintain proper protein homeostasis.

Pathways

EDEM1 contributes to the unfolded protein response (UPR) and the ERAD pathway. Both pathways are essential for managing protein folding and degradation within the cell. EDEM1 interacts with proteins like HERP and OS-9 during these processes helping with the recognition and processing of misfolded proteins. The correct functioning of these pathways is important for cellular adaptation to stress.

EDEM1's malfunction can link to conditions such as cystic fibrosis and neurodegenerative diseases. These diseases can arise due to defects in the clearance of misfolded proteins leading to their accumulation. EDEM1 interacts with proteins such as CFTR where improper folding is a known issue in cystic fibrosis. Its role in preventing the buildup of misfolded proteins makes it relevant to the pathology of these disorders.

Extracts misfolded glycoproteins, but not glycoproteins undergoing productive folding, from the calnexin cycle. It is directly involved in endoplasmic reticulum-associated degradation (ERAD) and targets misfolded glycoproteins for degradation in an N-glycan-independent manner, probably by forming a complex with SEL1L. It has low mannosidase activity, catalyzing mannose trimming from Man8GlcNAc2 to Man7GlcNAc2.
