补充信息

ECSIT also known as Evolutionarily Conserved Signaling Intermediate in Toll pathways is a mitochondrial protein involved in various cellular processes. It has a molecular mass of approximately 52 kDa and is prominently expressed in tissues with high metabolic activity such as the heart and skeletal muscles. ECSIT interacts with the mitochondrial membrane and is integral for mitochondrial function and biogenesis. Understanding the expression and function of ECSIT can provide insight into its role in cellular energy homeostasis.
Biological function summary

ECSIT influences several important cellular processes through its participation in the assembly of the mitochondrial complex I. It is part of a protein complex that includes NDUFAF1 and acts as a scaffold protein facilitating the proper assembly of complex I. ECSIT involvement is important for the maintenance of cellular respiration and energy production. Its role in complex I assembly highlights its importance in regulating mitochondrial activity and function.

Pathways

ECSIT acts in both mitochondrial biogenesis and the toll-like receptor (TLR) signaling pathway. In the TLR pathway ECSIT mediates interactions between TRAF6 and TAK1 contributing to the regulation of immune responses. It is an essential component linking mitochondrial function with immune signaling demonstrating its multi-functional capability. These pathways highlight the intricate network where ECSIT collaborates with proteins like TRAF6 and TAK1 to ensure proper immune response and energy metabolism.

ECSIT has associations with leigh syndrome and aggressive lung cancer. Its dysfunction in mitochondrial biogenesis can contribute to Leigh syndrome a severe neurological disorder. In lung cancer ECSIT involvement in TLR signaling affects cell proliferation and survival making it a protein of interest in cancer research. ECSIT's role in these conditions can involve interactions with mitochondrial proteins and signaling molecules like TRAF6 further illustrating its impact on human health.

靶点信息

Adapter protein that plays a role in different signaling pathways including TLRs and IL-1 pathways or innate antiviral induction signaling. Plays a role in the activation of NF-kappa-B by forming a signal complex with TRAF6 and TAK1/MAP3K7 to activate TAK1/MAP3K7 leading to activation of IKKs (PubMed : 25355951, PubMed : 31281713). Once ubiquitinated, interacts with the dissociated RELA and NFKB1 proteins and translocates to the nucleus where it induces NF-kappa-B-dependent gene expression (PubMed : 25355951). Plays a role in innate antiviral immune response by bridging the pattern recognition receptors RIGI and MDA5/IFIT1 to the MAVS complex at the mitochondrion (PubMed : 25228397). Promotes proteolytic activation of MAP3K1. Involved in the BMP signaling pathway. Required for normal embryonic development (By similarity).. As part of the MCIA complex, involved in the assembly of the mitochondrial complex I.
