Immunohistochemistry (PFA fixed) - Anti-Eagle antibody [8B11DH10] (AB201327)

Immunohistochemistry analysis in neuroblast nuclei from Drosophila stage 15 embryos. The cells were fixed in 4% formaldehyde for 25 minutes, permeabilized with 0.1% Triton X-100/PBS, and blocked in 0.1% Triton X-100/PBS, 2.5% Normal Goat Serum, 2.5% Normal Donkey Serum. The Eagle antibody (ab201327) was used at 3.8 μg/mL, overnight at 4°C. The secondary antibody used was Alexa488 conjugated goat anti-mouse at 1/400.

Eagle also known as ELK3 or Net is a nuclear protein that weighs approximately 44 kDa. It is a member of the ETS domain transcription factor family. Eagle primarily localizes in the nucleus and is widely expressed in various cell types with higher concentrations observed in the spleen and thymus. The protein shows a significant affinity for binding to DNA sequences and influences the transcriptional regulation of genes involved in cellular proliferation and differentiation.
Biological function summary

Eagle plays a significant role in the regulation of gene expression. It functions as part of a multiprotein complex involving other ETS domain family members. Eagle can act as both a transcriptional activator and repressor depending on the cellular context. Its regulatory activities are important in the processes of vascular development and maintenance of normal cellular function.

Pathways

Eagle is actively involved in the Ras-MAPK signaling pathway. This pathway is important for transmitting signals from the cell surface to the nucleus controlling cell cycle progression and modulating differentiation responses. Eagle interacts closely with other proteins in this pathway such as MEK and ERK influencing the transcriptional regulation downstream of the signaling cascade. Eagle's participation in this pathway reflects its ability to affect various cellular outcomes through modulating gene expression.

Eagle has been associated with several types of cancer including breast and colorectal cancer. Its dysregulation affects the transcriptional control of genes linked to uncontrolled cell proliferation. Eagle's interaction with the protein c-Fos in these cancers contributes to altered signaling pathways that favor oncogenesis. Understanding Eagle's role in these diseases highlights its potential as a target for therapeutic intervention in cancer research.
