EAG1 also known as Kv10.1 is a voltage-gated potassium channel with a molecular mass approximately 110 kDa. This protein plays a critical role in regulating membrane potential by allowing potassium ions to flow in response to voltage changes across the cell membrane. EAG1 is prominently expressed in the brain with presence noted in other tissues such as the heart and peripheral nervous system. Its activity influences the electrical excitability of neurons and has implications in cellular signaling.
Biological function summary

EAG1 is involved in key processes governing cellular excitability and proliferation. The channel contributes to setting the electrical properties of cells especially in neurons by controlling action potential dynamics. EAG1 does not typically form part of a complex but it can interact with other membrane proteins or signaling molecules modulating its activity. In certain contexts its aberrant expression can lead to altered cellular behaviors linking it to various cellular functional states.

Pathways

EAG1 engages in the regulation of the MAPK signaling pathway and is also significant in the PI3K/AKT pathway. These pathways are important for transmitting cellular signals that influence cell growth and survival. In the context of the MAPK pathway EAG1’s activity may interact with proteins such as B-Raf affecting downstream signaling events. Similarly in the PI3K/AKT pathway interactions can occur with proteins like AKT further integrating EAG1 within these essential frameworks.

EAG1 shows a connection to cancer and epilepsy. EAG1 overexpression has been documented in various cancers including breast and colon cancers where it may drive tumorigenic processes through its influence on cell proliferation and survival pathways. Moreover aberrant EAG1 function links to epilepsy potentially through its impact on neuronal excitability and neurotransmission. In these conditions EAG1 may interact with proteins such as p53 in cancer altering cell cycle control or with neurotransmitter receptors in epilepsy impacting neuronal networks.

Pore-forming (alpha) subunit of a voltage-gated delayed rectifier potassium channel that mediates outward-rectifying potassium currents which, on depolarization, reaches a steady-state level and do not inactivate (PubMed : 10880439, PubMed : 11943152, PubMed : 22732247, PubMed : 25420144, PubMed : 25556795, PubMed : 25915598, PubMed : 27005320, PubMed : 27325704, PubMed : 27618660, PubMed : 30149017, PubMed : 9738473). The activation kinetics depend on the prepulse potential and external divalent cation concentration (PubMed : 11943152). With negative prepulses, the current activation is delayed and slowed down several fold, whereas more positive prepulses speed up activation (PubMed : 11943152). The time course of activation is biphasic with a fast and a slowly activating current component (PubMed : 11943152). Activates at more positive membrane potentials and exhibit a steeper activation curve (PubMed : 11943152). Channel properties are modulated by subunit assembly (PubMed : 11943152). Mediates IK(NI) current in myoblasts (PubMed : 9738473). Involved in the regulation of cell proliferation and differentiation, in particular adipogenic and osteogenic differentiation in bone marrow-derived mesenchymal stem cells (MSCs) (PubMed : 23881642).
