Question

I used this DNA Ligase III antibody on proteins extracts from mouse fibroblasts and tissues. 40ug protein was loaded on SDS-PAGE gel and transfered using Trans-Blot machine onto PVDF membrane. Blocking was done using 5% Milk in TBS-T. 1:1000dilution of antibody was used for 24 hours at 4C followed by 1:5000 dilution of goat-anti rabbit antibody for 1 hour at Room Temperature. Development was done using Chemiluminescent substrate. All washes were done with TBS-T for atleast 10 min, three times. After this, I did not see any bands, neither for the fibroblasts nor for the tissue extracts. I am requesting a refund on this antibody or another replacement antibody that I can try on the extracts.

I am sorry that you have been experiencing difficulties with this antibody in WB. I have reviewed the protocol information you provided and I would like to make some suggestions that may improve your results with this antibody.


I noticed you are using mouse samples in your experiments. Have you also tested a positive control from human? Since we have not tested this antibody with mouse we cannot guarantee that it is compatible with this species. You would need to test human samples to confirm the antibody is not working.


I would like to recommend the following modifications:


Block using 5% BSA rather than milk - we routinely test our products with this reagent. Additionally, use 3% BSA as the antibody diluent for both the primary and secondary antibodies. Continue with the primary antibody incubation overnight at 4oC. Also confirm that the secondary antibody is working well with other antibodies.

Inquiry: This is in reference to my enquiry regarding ab97404. Thanks for getting back to me. I tried to follow the suggestions I received in the email from the technical team. Unfortunately, the blots are like the previous one that didn't work. The BSA only reduced background a little bit. I think it might a good idea to check with a different antibody.

Thank you for contacting us and for keeping us updated about the situation. I am sorry to hear that the suggestions from my colleague did not help improve the results.

At this time we have only tested ab97404 with human lysates, and though we predict cross-reactivity with mouse based on sequence homology, we are not able to guarantee the results in mouse. You can try the antibody in Western blot with human lysates, and if this also does not work I would be happy to send a replacement antibody.

For untested species (such as mouse in this case), we do have a testing discount program. We generally arrange this prior to purchase, but I will be happy to offer this program retroactively to you as well. You would simply need to submit an Abreview with the protocol and results seen with ab97404, and in exchange we will give you one free primary antibody. I would recommend trying ab125434, as this is the only DNA ligase III antibody that we have tested in WB with mouse lysates.
