DLC1 also known as Deleted in Liver Cancer 1 is a tumor suppressor protein with a mass of approximately 123 kilodaltons. It functions mechanically as a Rho GTPase-activating protein (RhoGAP) regulating the activity of Rho family GTPases. DLC1 is expressed in various tissues with significant expression in the liver lung and kidney. The protein helps maintain cytoskeletal integrity and cell stability by modulating actin dynamics and cellular contractility.
Biological function summary

The protein DLC1 plays an important role in cell migration proliferation and adhesion. It often forms part of a multi-protein complex with tensin proteins which helps link it to the actin cytoskeleton. Through its GAP activity DLC1 negatively regulates RhoA a small GTPase. This regulation affects cell shape and movement by controlling focal adhesion turnover and stress fiber formation essential processes in maintaining normal cellular architecture.

Pathways

DLC1 impacts cellular signaling related to the cell cycle and apoptosis. It is heavily involved in the regulation of the Rho/Rac signaling pathway which is critical for actin cytoskeleton organization and cellular dynamics. DLC1 interacts with proteins such as RhoA and Cdc42 within these pathways facilitating signal transduction that affects cellular responses to external stimuli and internal checkpoints ensuring proper cell division and programmed cell death.

DLC1 is closely related to various cancers including liver and breast cancer. Its tumor suppressor function is often lost or reduced in these conditions leading to unchecked cellular proliferation and metastasis. In the context of liver cancer DLC1 interacts with proteins such as PTEN influencing tumor growth and progression. Aberrations in DLC1 expression or activity can also correlate with other disorders involving disrupted cell signaling and cytoskeletal abnormalities.

Functions as a GTPase-activating protein for the small GTPases RHOA, RHOB, RHOC and CDC42, terminating their downstream signaling. This induces morphological changes and detachment through cytoskeletal reorganization, playing a critical role in biological processes such as cell migration and proliferation. Also functions in vivo as an activator of the phospholipase PLCD1. Active DLC1 increases cell migration velocity but reduces directionality. Required for growth factor-induced epithelial cell migration; in resting cells, interacts with TNS3 while PTEN interacts with the p85 regulatory subunit of the PI3K kinase complex but growth factor stimulation induces phosphorylation of TNS3 and PTEN, causing them to change their binding preference so that PTEN interacts with DLC1 and TNS3 interacts with p85 (PubMed : 26166433). The PTEN-DLC1 complex translocates to the posterior of migrating cells to activate RHOA while the TNS3-p85 complex translocates to the leading edge of migrating cells to promote RAC1 activation (PubMed : 26166433).
