As I am sure you are aware, the antibody purchased, Anti-Cytokeratin 5 + 6 antibody (ab17133) was ordered quite a long time ago. Almost a year prior to the report of the complaint. We would therefore not usually offer a replacement antibody as this requires that the complaint be made within 6 months of the purchase of the antibody to comply with the Abpromise guarantee. In this case I will offer a replacement vial of the antibody (ab17133) if your customer would like as a goodwill gesture.

I would however suggest that a few protocol alterations may be worthwhile in order to try to optimise the results observed.

1. I would suggest improving the specificity and strength of the detection by incubating with the primary antibody overnight as 4°C, not 37°C as is currently used. This antibody may not be suitable for use at elevated temperatures and we often find superior performance when used at lower temperatures for longer. This has been employed by the customer who has used this antibody previously and achieved the following staining:

2. You have mentioned that the antigen retireval has been optimised. How has this been performed? For this particular target or tissue? I would suggest trying the Citrate buffer currently used as well as Tris-EDTA (pH 9) under a few different times (3, 5, 10 and 20 minutes). This can often provide drastic differences in performance and optimisation may be required for each target you are interested in.

3. I can suggest to consider trying different times for antigen retreival. Has this already been tried?This can sometimes require some optimization.

4. Is the sedondary antibody and detection kit working well with other primary antibodies?
