Connexin 40.1 also known as GJA5 is a gap junction protein that facilitates direct cell-to-cell communication. This protein exhibits a mass of approximately 40 kDa and is commonly found in cardiac tissue as well as in vascular smooth muscle cells. Connexin 40.1 forms channels that allow ions and small molecules to pass between adjacent cells providing a path for the coordination of cellular activities. Its expression is mainly concentrated in the atrial tissues of the heart where its role in electrical coupling is critical for proper cardiac function.
Biological function summary

Gap junctions formed by Connexin 40.1 are essential for maintaining synchronized cardiac conduction. Connexin 40.1 interacts with other connexins to form heteromeric and heterotypic channels enhancing the complexity and selectivity of gap junctions. These channels participate in the propagation of action potentials across myocardial tissue contributing to rhythmic heart contractions. As part of these junctions Connexin 40.1 helps maintain cardiac stability and is pivotal in electrochemical coupling within the heart.

Pathways

Connexin 40.1 plays a role in the cardiac conduction system and the vasomotor regulation pathway. The protein cooperates closely with Connexin 43 and other connexins facilitating the propagation of electrical signals that drive myocardial contraction. In the heart Connexin 40.1 is part of the pathway responsible for facilitating atrial conduction ensuring efficient atrial impulse propagation. In vascular tissues it influences vasomotor tone by supporting cell communication necessary for smooth muscle contraction and relaxation.

Mutations or defects in Connexin 40.1 genes have ties to cardiovascular conditions like atrial fibrillation and familial atrial standstill. Such disorders disrupt normal cardiac rhythm and are associated with irregular heartbeat patterns. Connexin 43 can also be implicated in these disorders as both proteins often work in conjunction to maintain normal cardiac conduction. Studying these relationships enhances understanding of the mechanisms that underlie cardiac pathologies and aids in developing therapeutic interventions.
