Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-Collagen III antibody [FH-7A] (AB6310)
ab6310 at 1/600 diltuion staining preovulatory follicle and whole ovary tissue sections by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections). Antigens were retrieved by boiling with an antigen unmasking solution for 20 min at 120°C in an autoclave and then cooled down in water for 5 minutes. The tissue sections were formaldehyde fixed and incubated with the antibody for 1 hour. An alkaline phosphatase conjugated antibody was used as the secondary. The image shows a section of whole preovulatory follicle. Staining for collagen type III is seen in the theca interna cell layer. No staining in the granulosa cells.
This image was generated using the ascites version of the product.

Western blot - Anti-Collagen III antibody [FH-7A] (AB6310)
The protein can be used reduced + denaturated (band size ~130 kDa) or native (two bands ~ 130kDa and ~300kDa) The native protein gives a brighter signal.
PH of the protein lysate is low - a loading dye with bromphenolblue changes its colour. This does not effect the western blot.

Collagen III is a critical fibrillar collagen that plays a significant role in the structural integrity of various tissues, including the skin, lungs, and blood vessels. Alterations in Collagen III expression are associated with several diseases. For instance, mutations in the COL3A1 gene, which encodes Collagen III, can lead to Ehlers-Danlos syndrome type IV, a condition characterized by fragile blood vessels and organs. Additionally, abnormal Collagen III deposition is implicated in fibrotic diseases, such as liver cirrhosis and pulmonary fibrosis, where excessive collagen accumulation disrupts normal tissue architecture. Understanding Collagen III's role in these diseases is crucial for developing targeted therapies and improving patient outcomes.
Type III collagen, [a1(III)]3 ,is an approx. 300 kDa molecule, found predominantly in skin, blood vessels, liver, placenta, tongue, and thymus. Collagen type III forms cofibrils with type I and/or V collagens in a number of tissues of mesenchymal origin, such as skin, tendon, ligaments, and bone. This collagen type is involved, directly or indirectly in several genetic diseases, including Ehlers-Danlos type IV disease.

Biological function summary
Collagen type III contributes to maintaining the structural integrity of tissues. It forms a complex with collagen type I providing a scaffold that ensures mechanical strength and stability to tissues. This structural pairing of collagen type I and III highlights their cooperative function in supporting the extracellular matrix and regulating cell behavior in tissue repair and wound healing processes.
Pathways
Collagen type III is integral to the processes of tissue repair and fibrosis pathways. It interacts closely with proteins such as fibronectin and integrins contributing to the cellular adhesion and signaling pathways important for fibroblast function and migration. These interactions reflect its essential role in matrix assembly and stability influencing how cells respond to injury and repair damaged tissues.
