        Immunocytochemistry/ Immunofluorescence - Anti-Coilin antibody [Pdelta] (ab11822)
        ICC/IF image of ab11822 stained MCF-7 cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab11822, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-mouse IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

      Question

      Dear Sir/Madam, I recently purchased this antibody.My reason for purchasing was my requirement for an antibody specific to human and that does NOT cross-react with mouse. According to information prior to my purchase, and the datasheet accompanying the product, your anti-Coilin antibody should meet this requirement. Unfortunately, after twice attempting immunocytochemistry with this product, I have seen uniform reaction with mouse tissue. I am concerned about the specificity of this product. Please note, I am away on holiday for three weeks from tomorrow, but intend to re-address my experiment upon my return. If you have any suggestions concerning the use of this product I'd be happy to receive them. However, if I continue to get reactivity with mouse, then I intend to return the remainder of the product with a view to obtaining a refund. With thanks for your attention, Yours faithfully,

      Thank you for your enquiry. We would like to confirm that this antibody has been tested and characterized for Western blot and ICC application. It recognizes human Coilin but it does not cross-react with Mouse However we would like to emphasize the following: For immunohistochemistry, the primary antibody should be raised in a species as phylogenetically different as possible to the species that your sample was obtained. This is to avoid the secondary antibody cross-reacting with endogenous immunoglobulins in the sample, leading to erroneous results. For instance, this primary antibody used to detect a protein in a sample from a mouse should not be raised in mouse or rat. A primary antibody raised in rabbit would be a more appropriate choice. This is not really an issue with other immunological techniques such as Western blot application where samples do not contain endogenous immunoglobulin.
