Coatomer subunit delta also called COPD is a component of the COPI coatomer complex involved in membrane trafficking processes. This protein has a mass of approximately 50 kDa and is found in Golgi-associated transport systems. It interacts with other coatomer proteins to form a multi-subunit complex that assists in transporting molecules between Golgi compartments and from the Golgi to the endoplasmic reticulum. Its presence is typically noted in most eukaryotic cells contributing to its essential role in the cellular transport machinery.
Biological function summary

The coatomer subunit delta participates as a part of the COPI coatomer complex which is essential in vesicle formation. This complex stabilizes curvature in membranes to help form vesicles important for intracellular transport. It is key in the recycling of proteins and lipids to the endoplasmic reticulum maintaining homeostasis within the cell. The coatomer complex works with proteins such as ARF1 which activates the assembly of the coatomer complex onto membranes facilitating proper cargo selection and vesicle budding.

Pathways

The coatomer subunit delta is integral to vesicular trafficking pathways including the retrograde transport pathway from Golgi to the endoplasmic reticulum. It plays a significant role in the secretory pathway which is central to the transport of proteins and lipids. It has interactions with other coatomer subunits and accessory proteins ensuring efficient pathway function. It relates closely to proteins such as ARF1 and various cargo receptors which are essential for sorting and transport of cargo.

Improper functioning of the coatomer subunit delta can contribute to neurodegenerative conditions and certain types of cancer. Its disturbance may relate to Alzheimer’s disease where cellular transport mechanisms are disrupted leading to protein misfolding and aggregation. Additionally abnormalities in coatomer subunits are linked to some cancers potentially due to their influence on cell growth and proliferation. The interactions with proteins like ARF1 can amplify these effects highlighting the necessity of functional coatomer subunit delta for health maintenance.

靶点信息

Component of the coatomer, a cytosolic protein complex that binds to dilysine motifs and reversibly associates with Golgi non-clathrin-coated vesicles, which further mediate biosynthetic protein transport from the ER, via the Golgi up to the trans Golgi network. The coatomer complex is required for budding from Golgi membranes, and is essential for the retrograde Golgi-to-ER transport of dilysine-tagged proteins. In mammals, the coatomer can only be recruited by membranes associated to ADP-ribosylation factors (ARFs), which are small GTP-binding proteins; the complex also influences the Golgi structural integrity, as well as the processing, activity, and endocytic recycling of LDL receptors (By similarity).
