The protein known as Claudin 10 also referred to as CLDN10 is a member of the claudin family essential for formation and maintenance of tight junctions. Claudin 10 has a molecular weight of approximately 24 kDa. This protein is expressed widely across various tissues with significant levels in the kidney liver and pancreas. Its main role involves controlling the permeability of epithelial barriers contributing to paracellular transport regulation.
Biological function summary

The function of Claudin 10 includes its involvement in forming and maintaining tight junction strands within epithelial and endothelial cells. It participates in a complex with other claudins to create selectively permeable barriers that regulate ion transport. Claudin 10 plays a role in determining cation selectivity and permeability impacting various physiological processes such as electrolyte balance and fluid homeostasis. In kidney tubules it especially influences the reabsorption and secretion of ions.

Pathways

Claudin 10 interacts with signaling mechanisms that regulate epithelial tight junction integrity. One significant pathway is the Wnt signaling pathway which influences cell polarity and growth affecting tight junction dynamics. Claudin 10 also associates with proteins related to the Par3/Par6/aPKC complex within these pathways. Its function intertwines with those of other tight junction proteins such as ZO-1 and occludin forming a network vital for epithelial cell barrier function and tissue homeostasis.

Altered expression of Claudin 10 has links to kidney disorders such as hypermagnesemia with hypocalciuria due to its important role in magnesium reabsorption. Forms paracellular channels : polymerizes in tight junction strands with cation- and anion-selective channels through the strands, conveying epithelial permeability in a process known as paracellular tight junction permeability.. Isoform 1. Forms cation-selective paracellular channels. In sweat glands and in the thick ascending limb (TAL) of Henle's loop in kidney, it controls paracellular sodium permeability which is essential for proper sweat production and renal function (PubMed : 19383724, PubMed : 28686597, PubMed : 28771254, PubMed : 35650657, PubMed : 36008380).. Isoform 2. Forms anion-selective paracellular channels.
