Immunohistochemical staining and retrograde tracing of the EVN.VN : vestibular nucleus; G7n : genu of seventh cranial nerve (facial nerve); 6n : sixth cranial nerve nucleus (abducens nucleus). (A&C) Immunohistochemical staining for CGRP (n = 7) and ChAT (n = 4) respectively in transversely sectioned mouse brainstem. The EVN is located dorsolateral to genu of seventh nerve (box). Neurons in the abducens and vestibular nucleus were also labelled. (B&D) Higher power visualization of vestibular efferent nucleus. Arrowheads indicate CGRP and ChAT immuno-positive cells respectively. (E&F) Fluorogold injection into the posterior canal of ChAT : : tdTomato mouse strain under low and high power respectively (n = 4). Arrowheads indicate co-localization (yellow) of fluorogold (green) and tdTomato (red), confirming the location of vestibular efferent nucleus.

CCR2 is widely expressed in DRG neurons and displays upregulation following peripheral inflammation. a, b Typical examples and quantitative summary from dual immunofluorescence experiments showing CCR2 immunoreactivity IB4-binding non-peptidergic DRG neurons in the naïve and CFA-inflamed mice. c, d Immunofluorescence experiments and quantitative summary showing the coexistence of CCR2 immunoreactivity in CGRP-expressing peptidergic DRG neurons in naïve mice and its upregulation in CFA-inflamed mice. e, f Immunofluorescence staining and quantitative summary showing the colocalization of CCR2 immunoreactivity with NF200-positive large-diameter DRG neurons in naïve mice and no significant change in expression level after CFA inflammation. Note that in CFA-inflamed group, ipsilateral L3/L4 DRGs were obtained and analyzed at 24 h after intraplantar CFA injection. Arrows indicate the co-expressing cells. **P < 0.01. Two-Way ANOVA followed by post-hoc Bonferroni test (n = 4 mice/group). Data are expressed as mean ± SEM

Target Information
CGRP (Calcitonin Gene-Related Peptide) is a significant neuropeptide extensively studied in neuroscience. Its role is particularly noted in various neurodegenerative conditions, including Alzheimer's disease (AD). CGRP is involved in modulating inflammation and neuroprotection, making it a valuable marker for assessing neuroinflammatory responses. In AD, changes in CGRP expression can provide insights into the disease's progression and the associated inflammatory processes. This highlights CGRP's importance in understanding and potentially targeting neuroinflammation.

Biological function summary
CGRP contributes to numerous physiological processes including blood flow regulation body temperature control pain transmission and neurogenic inflammation. It acts independently and does not substantially form part of larger protein complexes in its primary activities. Through binding to the CGRP receptor a complex made up of the calcitonin receptor-like receptor (CLR) and receptor activity-modifying protein 1 (RAMP1) CGRP initiates downstream signaling that results in its various biological effects.
Pathways
Studies place CGRP within the neuropeptide signaling pathway and the vasodilation pathway. It significantly influences the cyclic AMP (cAMP) signaling pathway where it works collaboratively with other neuropeptides like substance P. The interaction with CLR and RAMP1 triggers the production of cAMP which enhances vasodilation and pain transmission processes in the body.
