    Important transcriptional activator in the regulation of genes involved in immune and inflammatory responses. Specifically binds to an IL-1 response element in the IL-6 gene. NF-IL6 also binds to regulatory regions of several acute-phase and cytokines genes. It probably plays a role in the regulation of acute-phase reaction, inflammation and hemopoiesis. The consensus recognition site is 5'-T[TG]NNGNAA[TG]-3'. Functions in brown adipose tissue (BAT) differentiation.

    We have not tested ab15050 in ChIP, therefore we cannot guarantee that it will work in this application. Since it does work for IP though, there is a good chance that it will be successful.

    Thank you for sending the mouse cell lysate once again. We have tried it in our western blot and I have attached the blots (although a bit dirty this time). What I believe we are seeing is one of the isoforms at just under 40 kDa, and it does appear in the cell lysate that there is a faint band at about 20 kDa. I was just wondering your thoughts on this. Perhaps in our samples we are getting a lot of unspecific binding and the smallest isoform is just not there?

    Thank you for getting back to me on this.

    I have put some red arrows next to what I believe are the correct size bands. It looks to me as though the correct bands are present in the mouse cell lysate (although faint) and that in your samples the 38 & 35kDa bands are present. It’s hard to say if the 20kDa band in just not present in your samples, or it present but a low levels. Is it possible that your samples would not contain the 20kDa form?

    To try and clean up the blot a little, I would recommend, unless you are already doing so in to use a 5% BSA block and also incubate your primary antibodies in the BSA as well. Also, increase the percentage of tween in your wash buffer to 0.1% and see if that helps to clean up the membrane.

    have ordered one of your CEBP beta antibodies, specifically:
    Anti-CEBP Beta antibody [1H7]
    ab15050
    On the datasheet it describes this antibody as being able to detect all isoforms of CEBPb including LIP (20 kDa) and LAP (35, 38 kDa). We have used this antibody in western blots with our own samples (mouse ovaries) and are slightly confused as to what isoforms we are detecting as they do not correspond to the sizes mentioned (I have included a sample of one of the blots). Is there a way to know which band corresponds to which isoform or a way to get protein that we could run as a positive control for this (either where LIP or LAP are perhaps overexpressed). This would allow us to see where the appropriate band might be?

    I have looked at the image you sent me and you are getting the correct banding pattern, but everything is 10-15kDa higher than it should be. So you could be really unlucky and the antibody is binding non-specifically (but still giving the triplicate banding), which I doubt, or the bands you are seeing are the correct bands, but for whatever reason are at a higher molecular weight.

    I think the best way to try and determine what is going on is for me to send you ab14632, 3T3-L1 (Mouse embryonic fibroblast - adipose like cell line) Nuclear Lysate.

    To send you the cell lysate, could you please either me your full shipping address, or send me the original order number for ab15050, so that I can get the address from there.

    I am interested in trying your C/EBPbeta antibody (ab15050). Is it possible to have a small sample? We currently use Santa Cruz's antibody and are very displeased. We need to find a company which has a high quality antibody to detect the smallest isoform LIP. We perform Western blots daily for this protein and if satisfied with your product we would purchase several vials yearly. I would appreciate any help with this. Thank you.
