      Flow Cytometry (Intracellular) - Anti-CD32B + CD32A antibody [AT10] - BSA free (ab41899)
      Overlay histogram showing K562 cells stained with ab41899 (red line). The cells were fixed with 80% methanol (5 min) and incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab41899, 1µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a positive result in 4% paraformaldehyde (10 min) fixed K562 cells used under the same conditions.

      Please note that Abcam do not have any data for use of this antibody on non-fixed cells. We welcome any customer feedback.

    Please could you advise as to whether the antibody below blocks the IgG Fc binding site when bound to the Fc receptor (CD32). I would like to use this product as a capture antibody in a sandwich ELISA using the following method. Anti-CD32 plate coating then+ CD32 then+ herceptin conjugated to detection reagent.

    According to Bunk S et al., this antibody completly blocksFcγRII (CD32) in human PBMCs.Please see the attachment.

    To our knowledge, ab41899 has not been tested in ELISA.     Thank you very much for your interest in our products. ab89191 (Anti-CD32 antibody [MM0270- 3L1]) from our catalogue was raised using CD32b extra cellular domain and reacts with both CD32a and CD32b. Unfortunately, it was not specifically tested if it reacts with CD32c. But since the CD32c and CD32b extra cellular domains are identical, we assume that it should also recognize CD32c. This antibody is tested and guaranteed to work in WB with human samples by our Abpromise guarantee. I am also happy to confirm that ab41899 (Anti-CD32 antibody [AT10]) binds to the extracellular region of CD32 and reacts with all isoforms of CD32, as described in Sandilands, G.P. et al. (2007) Immunol. 91: 204-211. Furthermore, clone AT10 has been shown to block CD32 and displace any antigen (IgG) that is bound to the receptor. This antibody is not tested and guaranteed for WB. I can confirm the following details which I have obtained from the laboratories: ab41899 CD32 antibody: Unfortunately, we do not have a specific protocol for the use of this product in blocking experiments, this application is supported based upon the literature. There are a number of articles which may be helpful to you, I have listed some examples below: Larsson M, Berge J, Johansson AG, Forsum U.     As far as I am aware, there has not been a study formally identifying the minimal epitoperecognised by this antibody. However, there is some information on the epitope recognised in the original paper describing the antibody: Greenman J, Tutt AL, George AJ, Pulford KA, Stevenson GT, Glennie MJ. Characterization of a new monoclonal anti-Fc gamma RII antibody, AT10, and its incorporation into a bispecific F(ab')2 derivative for recruitment of cytotoxic effectors. Mol Immunol. 1991 Nov;28(11):1243-54. Metes D, Ernst LK, Chambers WH, Sulica A, Herberman RB, Morel PA.
