Please see below for an email we have received from a customer;

I am writing with regards to your anti-Calcineurin antibody a3673 and the Abcam promise guarnatee.
This antibody has been used in our lab for many years at a concentration of 1:2000 in Western Blots to detect two bands of interest: The 61 kDa band, and the 48kDa cleavage product. We load 100ug of total protein and during the chemiluminescence exposure, we overexpose the more abundant 61 kDa band, to reveal the 48kDa claeavage product we are interested in.
In a comparison experiment, two seperate gels, loaded with identical samples of mouse brain homogenate, were transfered to two blots. One was probed with the old (effective) anti-Calcineurin antibody a3673, one with a new one purchased in August (lot number GR89521-1, 100ul).( PO 47096590 )
The new antibody completely failed to detect the presence of the 48kDa band (see attached scan). As this is the band we are interested in, and purchased the antibody only because it was in the past binding to this band, we would like to enquire about a replacement (a different lot number) or an alternative product you could offer us as part of your guarantee.

I can confirm that the Anti-Calcineurin A antibody (ab3673) is a ployclonal antibody. The affinity of these types of antibodies will change with different rounds of immunisation as well as different hosts. With different host even the epitopes of recognition might change. For that reason it is not unusual that with another lot the detection of the cleavage product might change.

We would be pleased to make an exception in this special case and offer you another lot of this product to try it with your samples. Please note that we cannot guarantee that this lot will detect the cleavage product the same as the first lot you purchased.

You're right, this will be immunocytochemistry - apologies for the error.
The cells are rat hippocampal neurons, dissected at E18, dissociated in trypsin, then grown for ˜10d on laminin- and PDL-coated coverslips, then fixed in paraformaldehyde (normally 4% for 20min, but we may try weaker fixation regimes to try and improve staining if necessary).

We'll be using this antibody for immunohistochemistry in dissociated hippocampal neurons. We have a great deal of experience in immuno labelling in these cells, and we know that particular treatments applied to the neurons should change the intensity and/or distribution of labelling for calcineurin. We also have numerous controls to either overexpress or block calcineurin in these neurons, so if we see staining changes in treatment groups we can be very sure of their authenticity.

Could you please confirm if the cells are human or murine origin?
As I understand you wish to test this antibody in immunocytochemistry and not immunohistochemistry. What method will be used to dissociate the hippocampal neurons and for fixating them? Enzymatic or other techniques?
It is important for us and for our customer to distinguish between these two applications Immunocytochemistry (ICC) and immunohistochemistry (IHC-P or IHC-Fr).
