CACNA1H also known as Cav3.2 or T-type calcium channel alpha-1H subunit functions as a voltage-dependent calcium channel. The protein plays a role in the influx of calcium ions into cells. It has a molecular mass of approximately 260.7 kDa. CACNA1H expresses most abundantly in the central nervous system although you can also find it in the heart and various other tissues. This channel is part of the larger Cav3 family of T-type calcium channels which includes other subunits like Cav3.1 and Cav3.3.
Biological function summary

This calcium channel regulates numerous cellular processes through its role in calcium signaling. It does not form part of a complex but still interacts closely with various signaling molecules to influence activities such as neurotransmitter release and muscle contraction. By modulating membrane potentials and synaptic transmission CACNA1H plays a significant direct role in neuronal excitability and rhythmic activities.

Pathways

CACNA1H interacts within the calcium signaling and neurotransmission pathways. In these pathways it regulates calcium ion homeostasis and neuronal excitability working alongside other channels like L-type and N-type calcium channels. CACNA1H affects the excitation-contraction coupling critical for smooth muscle function interlinking with proteins such as calmodulin that respond to changes in calcium ion concentration.

CACNA1H has been linked to epilepsy and neuropathic pain. In epilepsy mutations in CACNA1H can disrupt normal calcium flow affecting neuronal firing patterns and contributing to seizures. Its interaction with other ion channels like SCN1A further influences epileptic manifestations. Mutations may also contribute to neuropathic pain by altering pain threshold and response through its role in pain pathway modifications.

Voltage-sensitive calcium channel that gives rise to T-type calcium currents. T-type calcium channels belong to the 'low-voltage activated (LVA)' group. A particularity of this type of channel is an opening at quite negative potentials, and a voltage-dependent inactivation (PubMed : 27149520, PubMed : 9670923, PubMed : 9930755). T-type channels serve pacemaking functions in both central neurons and cardiac nodal cells and support calcium signaling in secretory cells and vascular smooth muscle (Probable). They may also be involved in the modulation of firing patterns of neurons (PubMed : 15048902). In the adrenal zona glomerulosa, participates in the signaling pathway leading to aldosterone production in response to either AGT/angiotensin II, or hyperkalemia (PubMed : 25907736, PubMed : 27729216).
