      Voltage-sensitive calcium channels (VSCC) mediate the entry of calcium ions into excitable cells and are also involved in a variety of calcium-dependent processes, including muscle contraction, hormone or neurotransmitter release, gene expression, cell motility, cell division and cell death. The isoform alpha-1C gives rise to L-type calcium currents. Long-lasting (L-type) calcium channels belong to the 'high-voltage activated' (HVA) group. They are blocked by dihydropyridines (DHP), phenylalkylamines, benzothiazepines, and by omega-agatoxin-IIIA (omega-Aga-IIIA). They are however insensitive to omega-conotoxin-GVIA (omega-CTx-GVIA) and omega-agatoxin-IVA (omega-Aga-IVA). Calcium channels containing the alpha-1C subunit play an important role in excitation-contraction coupling in the heart. The various isoforms display marked differences in the sensitivity to DHP compounds. Binding of calmodulin or CABP1 at the same regulatory sites results in an opposit effects on the channel function.

      Question

      Thank you for your former advice and replacement antibody for DHPR alpha 1 (aka CaV 1.2 alpha 1).

      Now we have tried Ab 81980 with our human heart samples, but unfortunately it does not seem to work either, as it gives some aspecific band with a smaller MW than expected and no band occurs at the expected MW range.

      I attach a ppt slide with the result we have got.

      In the meanwhile, we have tried another antibody and that one works fine, the band coming at the right MW and control peptide blocking showed that it was a specific band. We are going to use that antibody for our further studies.

      I would like to ask if we can still get a refund or credit which we can spend purhasing some other (not CaV 1.2) antibody?

      Out of either ab81980 or ab8109, I would recommend ab81980, as we have a western blot image for that one on our website. We do not have any more data for either antibody, although both are guaranteed to work in western blot on human samples.

      Thank you for your suggestions in order to make ab84814 work better. We have tried the suggestions, however, we still have a very strong background, no specific signal and lots of aspecific bands.

      I attach a word file which details the protocol which we routinely use and which is working with all the other antibodies, and also include the modifications you suggested last time.
      I also inserted a blot image we have got when following your advice.
      I would like to ask whether you have any more suggestions about what and how to modify in our protocol, or we would be happy to try another anti-DHPR antibody, like ab81980 or ab81095.

      I am sorry that the protocol tips did not prove to be successful. Under our Abpromise we do guarantee this antibody for 6 months to work in western blot on human samples. Since we have not been able to resolve this issue for you, then I would be happy to send you a replacement antibody as requested..
      To do that, I need to have your original order information. I believe I have found that information, could you confirm that the product was ordered on 9th Nov 2011 by Bethann Gabris. If this information is incorrect, could you send me the correct information? Also, I can send you either ab81980 or ab81095, just let me know which one you would like to receive.

      to the mail below, we ordered and tried "ab 84814" to perform western blot on human heart samples. We are rather disappointed as the antibody does give the specific band around 200 kDa, but it also gives some very heterogeneous background which makes it impossible to get clear image of the bands and analyse band intensity.    I attach a file to show the background I mean, so far we tried it 4 times with 3 different dilutions. Out of the 4, one blot was somewhat usable, the other 3 were unusable. I gave some information on our western blot protocol, if you need more details, please ask and I will write more. However, we are working with several antibodies and we never get this heterogeneous background with any of the others, so we think the problem is not with the protocol but with the antibody.   Could you please tell us what to do differently, or can we try either another batch of this, or a different CaV 1.2 alpha (=DHPR alpha 1) antibody?

      Thank you for contacting Abcam. I am sorry that you are having problems with ab84814 in western blot. I have looked over the images and protocol information that you sent and there are a few protocol changes that I would suggest that may help to reduce the background that you are seeing: 1 - Switch to a 5% BSA block rather than 5% Milk, as BSA is a little better a blocking non-specific binding. 2 - Increase the number of wash steps you are doing (up 5 washes, 5 minutes each) and add 0.05% Tween 20 to the wash buffer, if you have not done so already. 3 - After the secondary incubation, wash the membrane for 30 minutes in a 0.5M NaCl. Then rinse the membrane thoroughly in dH20 and develop as normal. If these protocol tips do not work then I would be happy to send you a replacement antibody, as under our Abpromise we do guarantee this antibody to work in western blot on human samples. I look forward to your reply and helping you resolve these issues.

      We are looking for a CaV 1.2 (aka DHPR alpha 1, aka CACNA1C) antibody suitable for western blotting on human heart total protein samples.    ab81980 seems the most suitable and the blot provided has no aspecific bands, however, ab84814 has more references and also predicted to react with human samples. Could you tell us which one has the better chance to react with CaV 1.2 without giving aspecific bands?   I also would like to ask about your guarantee policy, is that valid for any of these antibodies? Does it mean we can get our money refunded if the antibody does not perform well, or it means we can order some other antibody?

Under our Abpromise we guarantee all of our antibodies to work as stated on the datasheet, if they do work as stated and we cannot offer any assistance to resolve the matter, then we would either replace the antibody or give a refund. Both ab81980 and ab84814 are guaranteed to work in western blot on human samples. I would recommended ab84814, even though the western blot image may not be the cleanest.
