CABP or Calcium-Binding Protein is a protein that modulates calcium signaling pathways. Alternate names include calbindin and calretinin though these may refer to specific types within the broader CABP family. CABP proteins vary in mass generally ranging from 10 to 50 kDa depending on the specific family member. These proteins are mainly expressed in the nervous system where they play an important role in regulating intracellular calcium levels. They also appear in other tissues like the heart and kidneys where they contribute to various cellular functions.
Biological function summary

The regulation of calcium ions by CABP influences several processes including neurotransmitter release muscle contraction and cell division. CABP proteins can be parts of larger complexes where they interact with other calcium-binding proteins to modulate calcium-dependent signaling mechanisms. Their function in the regulation of calcium homeostasis suggests a direct involvement in maintaining normal cellular functions especially in neurons.

Pathways

CABP proteins play integral roles in calcium signaling and related pathways. They are critical in modulating the response of cells to calcium transients by interacting with proteins such as calmodulin and calcineurin. CABP's role in these calcium-mediated pathways ensures proper synaptic activity and muscle response and is important for the plasticity of neuronal connections critical for learning and memory.

The dysfunction of CABP has links to neurodegenerative disorders and cardiac diseases. Alterations in CABP expression can contribute to conditions like Alzheimer's disease where calcium signaling becomes dysregulated. In addition aberrant CABP function may also play a role in cardiac hypertrophy due to imbalanced calcium handling often involving calmodulin and other calcium-dependent molecules. Understanding CABP's involvement in these conditions could provide insights into potential therapeutic strategies.

靶点信息

Modulates calcium-dependent activity of inositol 1,4,5-triphosphate receptors (ITPRs) (PubMed : 14570872). Inhibits agonist-induced intracellular calcium signaling (PubMed : 15980432). Enhances inactivation and does not support calcium-dependent facilitation of voltage-dependent P/Q-type calcium channels (PubMed : 11865310). Causes calcium-dependent facilitation and inhibits inactivation of L-type calcium channels by binding to the same sites as calmodulin in the C-terminal domain of CACNA1C, but has an opposite effect on channel function (PubMed : 15140941). Suppresses the calcium-dependent inactivation of CACNA1D (By similarity). Inhibits TRPC5 channels (PubMed : 15895247). Prevents NMDA receptor-induced cellular degeneration. Required for the normal transfer of light signals through the retina (By similarity).
