ab55811 staining C5b-9 in human liver.
Left panel : with primary antibody at 4 ug/ml. Right panel : isotype control.
Sections were stained using an automated system (DAKO Autostainer Plus ), at room temperature : sections were rehydrated and antigen retrieved with the Dako 3 in 1 AR buffers citrate pH6.1 in a DAKO PT link. Slides were peroxidase blocked in 3% H2O2 in methanol for 10 mins. They were then blocked with Dako Protein block for 10 minutes (containing casein 0.25% in PBS) then incubated with primary antibody for 20 min and detected with Dako envision flex amplification kit for 30 minutes. Colorimetric detection was completed with Diaminobenzidine for 5 minutes. Slides were counterstained with Haematoxylin and coverslipped under DePeX. Please note that for manual staining we recommend to optimize the primary antibody concentration and incubation time (overnight incubation), and amplification may be required.

Immunohistochemical analysis of paraffin-embedded Human placenta tissue labeling C5b-9 with ab55811 at 1/100 dilution (24h at 4°C ) followed by Goat anti Rabbit Alexa Fluor® 488 at 1/1000 dilution (Green).

Red channel corresponds to Evans Blue and white channel is the fluorescence obtained from nuclear counterstain DAPI staining.

ICC/IF image of ab55811 stained HeLa cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab55811, 1μg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43μM.

C5b-9 also known as the membrane attack complex (MAC) serves as a critical component in the immune system's defense mechanism. Composed of proteins C5b C6 C7 C8 and multiple units of C9 this complex forms on the surface of pathogen membranes. It typically weighs approximately 900 kDa. The MAC largely expresses on cell membranes during immune responses and primarily targets bacterial pathogens by creating pores that lead to cell lysis.
Biological function summary

C5b-9 functions as part of the larger complement system which aids in innate immunity. The process begins once C5b initiates the assembly of the complex by binding to C6 C7 C8 and finally incorporating C9 molecules. These form a pore-like structure leading to increased permeability of the target cell membrane resulting in the disruption and elimination of the pathogen.

Pathways

C5b-9 plays a critical role in the complement cascade an essential pathway in the immune system. This cascade consists of several steps including the alternative and classical pathways. C5b-9 is related to the protein C3 through these pathways which upon activation helps facilitate C5 activation. This connection highlights its importance in promoting pathogen clearance.

C5b-9 has associations with conditions such as paroxysmal nocturnal hemoglobinuria (PNH) and atypical hemolytic uremic syndrome (aHUS). In PNH excessive formation of this complex on red blood cells leads to their destruction contributing to hemolysis. Similarly in aHUS dysregulation of the complement system including C5b-9 causes damage to endothelial cells. This complex connects with proteins like factor H and CD59 whose dysfunction or deficiency can lead to or exacerbate these diseases.
