Immunohistochemical analysis of Human tonsil tissue labeling C4d using ab183311 at a 1/100 dilution.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of Human tonsil tissue. The section was incubated with ab183311 at 1 : 100 dilution for 30 mins at room temperature. Heat-mediated antigen retrieval was performed with Tris-EDTA buffer (pH 9.0, epitope retrieval solution 2) for 20 mins. Rabbit specific IHC polymer detection kit HRP/DAB (ab209101) was used as the secondary antibody. A secondary antibody-only negative control was also performed. Hematoxylin was used as a counterstain. Positive staining was observed in the germinal centre of human tonsil performed on a Leica Biosystems BOND® RX instrument.

Immunohistochemical analysis of formalin fixed paraffin embedded human tonsil labelling C4d with ab183311 at a concentration of 1µg/ml. The immunostaining was performed on a Ventana DISCOVERY ULTRA (Roche Tissue Diagnostics) instrument with a OptiView DAB IHC Detection Kit. Heat mediated antigen retrieval was performed with DISCOVERY cell conditioning solution (CC1) 100°C, pH8.5 for 32mins.
ab183311 Anti-C4d antibody [SP91] - C-terminal was incubated for 16mins at 37°C. Sections were counterstained with Hematoxylin II. Image inset shows absence of staining in secondary antibody only control.
Customers are encouraged to optimise antigen retrieval conditions, antibody concentration, incubation times and temperature for best results in their own IHC assay workflow (automated and manual)

Biological function summary
The C4d protein acts as a marker for complement activation and is part of the larger complement cascade. C4d is primarily associated with the classical pathway and serves as an indicator of complement-mediated cell injury. The degradation of C4 into C4d is usually seen in immune responses where antibodies recognize antigens triggering sequence activation in the complement pathway. C4d's role as a marker facilitates the differentiation between antibody-mediated injury and other types of tissue damage.
Pathways
This complement protein gets involved in the classical pathway of the complement system which plays a critical role in innate and adaptive immunity. It is related to other proteins like C3 and C1q which are additional components within this pathway. These interactions are significant for the clearance of pathogens and also assist in enhancing the humoral immune response by promoting the deposition of complement on pathogenic surfaces.
